摘  要:目的采用超高效液相色谱串联四极杆飞行时间质谱(UPLC/Q-TOF MS)对生何首乌药材的化学成分进行分析及鉴定。方法采用ACQUITY UPLCTM BEH C18色谱柱(2.1 mm×50 mm×1.7μm),以乙腈-0.1%甲酸水溶液为流动相进行梯度洗脱,流速为0.4 mL/min,进样量为5μL,柱温为30℃,ESI源负离子模式下采集数据,根据Masslynx4.1软件分析结果并结合文献报道与数据库信息对比,对其中的化学成分进行有效辨别。结果鉴定了生何首乌药材中的22种化学成分,包括蒽醌类化合物7个,二苯乙烯苷类化合物5个,黄酮类化合物3个,有机酸2个,其他类化合物5个;其中2种二聚体化学成分(峰18,tR=12.771 min,m/z=757.1793;峰22,tR=20.997 min,m/z=523.1379)为首次从生何首乌药材中发现。结论建立了一种快速、有效鉴定生何首乌药材化学成分的UPLC/Q-TOF MS分析方法。Objective To analyze and identify the chemical components of raw Polygoni Multiflori Radix.by UPLC/Q-TOF MS.Methods The separation was performed on the ACQUITY UPLCTMTMBEH C18 chromatographic column(2.1 mm×50 mm,1.7μm),acetonitrile-0.1%formic acid aqueous solution was used as mobile phase in gradient elution with the flow rate of 0.4 mL/min,injection of 5μL and column temperature of 30℃.Data was collected through ESI source negative ion mode.The results were analyzed by Masslynx4.1 software and compared with the literature reported and database information to identify the chemical components effectively.Results 22 chemical components of Polygoni Multiflori Radix were identified,including 7 anthraquinones,5 distyrene glycosides,3 flavonoids,2 organic acids and 5 other compounds.Two dimer chemical components(peak 18,tR=12.771 min,m/z=757.1793;peak 22,tR=20.997 min,m/z=523.1379)were found in the chemical composition analysis of raw Polygoni Multiflori Radix for the first time.Conclusion A UPLC/Q-TOF MS analysis method has been established to rapidly and effectively identify the chemical components of raw Polygoni Multiflori Radix.
