HPLC法同时测定益肺清解汤中毛蕊异黄酮葡萄糖苷和绿原酸的含量

摘　　要：目的:建立一种快速、准确和实用的HPLC方法,用于同时测定益肺清解汤中毛蕊异黄酮葡萄糖苷和绿原酸的含量。方法:采用XterraRP18(5μm,4.6 mm×250 mm)色谱柱,以乙腈(A)-0.1%甲酸溶液(B)为流动相梯度洗脱,流速1.2 m L·min^-1,柱温40℃,检测波长为320 nm。结果:毛蕊异黄酮葡萄糖苷和绿原酸的进样质量分别在0.023~0.463μg(r=0.9999)、0.042~0.836μg(r=0.9998)范围内,与产生的峰面积线性关系良好;平均回收率分别为101.52%(RSD=2.56%)、100.39%(RSD=1.15%)。结论:本方法可作为我院中药成方制剂益肺清解汤中毛蕊异黄酮葡萄糖苷和绿原酸含量同时测定的方法,适用于该制剂的质量控制。Objective To establish a rapid,accurate and feasible HPLC method for simultaneous determination of calycosin-7-glucoside and chlorogenic acid in Yifeiqingjie soup.Methods The separation was carried out on a XterraRP18(5μm,4.6 mm×250 mm)column,the mobile phase consisting of acetonirtile(A)-0.1%formic acid solution(B)at a flow rate of1.2 m L·min^-1,with the column temperature of 40℃,the detection wavelength at 320 nm.Results The calibration curve was liner over the range of 0.023~0.463μg(r=0.9999)for alycosin-7-glucoside;0.042~0.836μg(r=0.9998)for chlorogenic.The average recovery of the two components was 101.52%(RSD=2.56%),100.39%(RSD=1.15%)respectively.Conclusion The method can be used for simultaneous determination of calycosin-7-glucoside and chlorogenic acid in Yifeiqingjie soup by our hospital traditional Chinese patent medicines,and it is applicative applicable for quality control of it.
