摘 要:以大兴安岭重度火烧迹地不同恢复年限落叶松人工林为研究对象,采用固体平板法、操作分类单元(OTU)聚类及注释方法,分析大兴安岭火烧迹地在植被恢复过程中土壤解磷微生物的数量、种群及活性。结果表明:天然次生林土壤解磷微生物的解磷效果最好,恢复23、8年的落叶松人工林土壤解磷微生物的解磷效果次之,恢复18年的落叶松人工林土壤解磷微生物的解磷效果较差;在不同解磷微生物中,无机磷溶磷菌的数量少于有机磷解磷菌的数量。对菌株通过序列拼接、过滤和去嵌合体后,得到优化序列,然后进行OTU聚类及注释。依据聚类结果,可以进行α多样性分析、β多样性分析;依据注释结果,可得到各水平的分类信息,从而进行样本组成及样本间群落结构差异相关分析。利用OTU聚类注释分析结果表明,在大兴安岭火烧迹地不同恢复年限的落叶松人工林土壤中包含14种不同科属的解磷细菌;对细菌的丰度分析表明,不同恢复年限的落叶松人工林的根际土,细菌种类和丰度相似,但非根际土之间解磷细菌的种类和丰度差异较大。Larch plantations of different restoration years were studied in the severely burned area in Daxing’an Mountains area in China. Solid plate method and OTU clustering and annotation method were applied to analyze the number, population and activity of soil phosphate-resolving microorganisms during the restoration of vegetation in the burned area. The natural secondary forest soil phosphate-resolving microorganisms represented the best phosphorus-resolving effect, 23- and 8-year-old larch plantation soils showed inferior effects, and 18-year-old larch plantation soils recovered the least amounts. The abundance of organophosphate-dissolving bacteria was greater than phosphate-dissolving bacteria. OTU clustering and annotation were conducted in the condition of optimized sequence through spliced, filtered and removing chimeras. Alpha diversity and Beta diversity analysis were realized with clustering results, and correlation analysis of sample compositions and community structure differences based on classification information at various levels with the annotation results were performed. Fourteen kinds of phosphate-resolving bacteria in different families and genera were found. The species and abundance of bacteria in rhizosphere soil of larch plantation with different recovery years were similar;nevertheless, the abundance of non-rhizosphere soil varied greatly.
