摘 要:为了明确紫穗槐(Amorpha fruticosa)叶斑病病原菌种类,采用常规组织分离法分离病原菌,依据柯赫氏法则对病原菌进行致病力测定,结合分子生物学技术及形态学鉴定确定病原菌分类地位,采用生长速率法测定了病菌对13种杀菌剂的敏感性。结果表明:引起紫穗槐叶斑病的致病菌为互隔交链孢(Alternaria alternata);适合菌丝生长的碳、氮源是葡萄糖和硝酸钠,适合孢子萌发的碳、氮源是蔗糖、硝酸钠和硝酸钙;适合菌丝生长和孢子萌发的温度分别是25、30℃;中性(pH=7)适合菌丝生长和孢子萌发;12 h-12 h光暗交替处理适合菌丝的生长;24 h全黑暗条件适合孢子萌发。病原菌菌丝对质量分数25%丙环唑EC、质量分数10%苯醚甲环唑WG敏感性较高,EC 50分别为84.97、95.99 mg·L^-1,其次为质量分数8%氟硅唑ME、质量分数10%IR9792 SC、125 g·L^-1氟环唑SC、250 g·L^-1啶氧菌酯SC,EC 50分别为264.11、290.33、305.9、350.82 mg·L^-1。To identify the pathogen of leaf spot on Amorpha fruteicosa for controlling the leaf spot.The isolated pathogens were tested for pathogenicity and the classification of the pathogens was determined by molecular techniques and morphological identification.The results showed the pathogen was Alter ariaalternata.The suitable carbon source for mycelium growth was Glucose,and NaNO 3 was the best nitrogen source.Forspore germination,the highest rate was on the sucrose,NaNO 3 and Ca(NO3)2 source.The best temperature for mycelium growth was 30℃and spore germination was 25℃.Both of spore germination and mycelium growth were at pH=7.The dark culture was the most suitable for spore germination and 12 h-12 h hight-dark was more favorable for mycelium growth.Using mycelium growth rate method to test the pathogen susceptibility on 13 fungicides.The pathogenic mycelium was sensitive to 25%propiconazole EC,10%difenoconazole WG,and EC 50 were 84.97 and 95.99 mg·L^-1,followed by 8%flusilazole ME,10%IR9792SC,125 g·L^-1 epoxiconazole SC,250 g·L^-1 picoxystrobin SC,and EC 50 were 264.11,290.33,305.9 and 350.82 mg·L^-1,respectively.
