摘 要:目的探讨黄芪、莪术配伍联合顺铂(DDP)对人肝癌裸鼠CD147、iNOS表达的影响。方法构建人肝癌裸鼠原位移植瘤模型。成瘤后,将其随机分为8组:模型对照组、阳性对照组(DDP)、黄芪莪术配伍高剂量组(H)、黄芪莪术配伍中剂量组(M)、黄芪莪术配伍低剂量组(L)、黄芪莪术配伍高剂量+顺铂组(H+DDP)、黄芪莪术配伍中剂量+顺铂组(M+DDP)、黄芪莪术配伍低剂量+顺铂组(L+DDP),每组8只,给药15d,免疫组织化学染色及Real-time RT-PCR检测肿瘤组织中CD147、iNOS蛋白及mRNA的表达。结果与模型对照组相比,H、M、H+DDP、M+DDP、L+DDP组CD147、iNOS蛋白及mRNA表达均显著下降(P<0.05或P<0.01),并与剂量呈一定的正相关性。与DDP组相比,H+DDP组CD147、iNOS蛋白表达降低(P<0.05或P<0.01)。结论黄芪、莪术配伍可能是通过下调CD147、iNOS蛋白及mRNA表达对肿瘤新生血管生成产生影响。Objective To study the effects on expression of CD147, iNOS in nude mice of human hepatocarcinoma transplantation model by compatibility of Radix Astragali and Curcuma and combination with cisplatin(DDP). Methods Human hepatocarcinoma cells HepG2 were transplanted into nude mice to form the study model, and the modeled mice were divided at random into 8 groups: model group, cisplatin(DDP)group(DDP), high dosage of Radix Astragali and Curcuma(H), intermediate dosage of Radix Astragali and Curcuma(M), low dosage of Radix Astragali and Curcuma(L), high dosage and cisplatin group(H+DDP), intermediate dosage and cisplatin group(M+DDP), low dosage and cisplatin group(L+DDP),8 in each group. The expression of CD147, iNOS protein and mRNA in tumor tissues were detected by the immunohistochemical staining technique and Real-time RT-PCR. Results Compared with the model group, the expression of CD147, iNOS protein and mRNA decreased significantly(P<0.05 or P<0.01) in H, M, H+DDP, M+DDP and L+DDP group. It was positively correlated with the dose. Compared with DDP group, the expression of CD147, iNOS protein obviously decreased(P<0.05 or P<0.01) in H+DDP group. Conclusion Compatibility of radix astragali and curcuma may affect tumor angiogenesis by down-regulating the expression of CD147, iNOS protein and mRNA.
