摘 要:目的建立超高效液相与质谱联用(UPLC-MS)方法测定藜芦与不同比例人参配伍前后主要生物碱经Caco-2细胞吸收转运后的变化情况,从吸收转运的角度分析人参"叛"藜芦的机制。方法液质条件为Acquity UPLC BEH C18(1.7μm, 2.1 mm×50 mm)色谱柱,柱温为35℃,流动相为乙腈(A)和0.1%甲酸水溶液(v/v),梯度洗脱;3200 QTRAPTM串联四级杆-线性离子阱质谱仪,采用电喷雾电离源,电喷雾电压为5.0 kV,透镜管电压为90 V,金属毛细管电压为40 V、温度为250℃。结果正离子扫描模式下共鉴定出8种藜芦生物碱,比较藜芦加入不同比例人参后几种藜芦生物碱峰面积的变化情况,发现大量人参的加入使藜芦生物碱的溶出增加,从而增加其毒性作用。从吸收转运角度分析,藜芦与一定比例的人参配伍后,藜芦生物碱的小肠吸收度升高,说明人参可促进藜芦生物碱的体内吸收。结论该研究结果从化学成分含量变化和吸收转运的角度再次验证了中药"十八反"中人参"叛"藜芦的配伍禁忌。Objective To establish a UPLC-MS method for the determination of the transport changes of the main alkaloids from Veratrum nigrum after the combination with Panax ginseng by the Caco-2 monolayer model. The mechanism of Panax ginseng "Insecticides" Veratrum nigrum could be studied from the perspective of absorption and transport. Methods Separation was performed on Acquity UPLC BEH C18 column(1.7 mm, 2.1 x 50 mm) and the column temperature was maintained at 35 °C.The mobile phase consisted of acetonitrile(A)and 0.1 % formic acid aqueous solution(B) with gradient elution. 3200 QTRAPTM Mass Spectrometer with ESI ionization source was set with electric spray fog voltage of 5.0 kV, metal capillary voltage of 40 V, lens tube voltage of 90 V, and the metal capillary temperature was 250 ℃. Results In the positive ion scanning model, 8 kinds of alkaloids were identified. And the changes of the peak area of several kinds of alkaloids were compared. It was found that the addition of Panax ginseng increased the dissolution of alkaloids and increased their toxicity.From the perspective of transport, the intestinal absorption of alkaloids increased after the combination with Panax ginseng. It showed that Panax ginseng could promote the absorption of the alkaloids. Conclusion The results of this study demonstrated once again the contraindication principle of Veratrum nigrum and Panax ginseng in the the theory of traditional Chinese medicine.
