摘  要:目的建立HPLC法同时测定肉苁蓉属药材中松果菊苷、金石蚕苷、毛蕊花糖苷、异毛蕊花糖苷、2′-乙酰金石蚕苷和2′-乙酰毛蕊花糖苷含量的方法。方法采用岛津Inertsil C18色谱柱(4.6 mm×250 mm,5.0μm),以乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱。流速为1.0 mL·min-1,柱温20℃,检测波长为330 nm。结果松果菊苷、金石蚕苷、毛蕊花糖苷、异毛蕊花糖苷、2′-乙酰金石蚕苷和2′-乙酰毛蕊花糖苷6个成分的进样量分别在0.240~2.400μg (r=0.9993),0.300~3.000μg (r=0.9998),0.072~0.720μg (r=0.9994),0.006~0.060μg(r=0.9991),0.150~1.500μg (r=0.9999),0.030~0.300μg (r=0.9993)线性关系良好;平均回收率(n=3)在98.81%~101.52%之间,RSD的范围为0.3%~2.8%。含量测定结果表明,肉苁蓉和管花肉苁蓉中含量最高的成分依次为松果菊苷、毛蕊花糖苷、异毛蕊花糖苷;肉苁蓉中均含有量较大的2′-乙酰毛蕊花糖苷;而管花肉苁蓉中检测不到;二者均不含金石蚕苷和2′-乙酰金石蚕苷。盐生肉苁蓉主要的成分为松果菊苷、毛蕊花糖苷、2′-乙酰毛蕊花糖苷,还有一定量的金石蚕苷。沙苁蓉的特征性成分为金石蚕苷和2′-乙酰金石蚕苷,毛蕊花糖苷和2′-乙酰毛蕊花糖苷含量极低,松果菊苷检测不到。结论建立的HPLC方法可把肉苁蓉属4种药材明确区分开来,方法简便、准确且重复性好,可望为肉苁蓉药材和饮片的质量评价和有效控制提供科学依据。Objective To establish an objective and reasonable HPLC method for simultaneous determination of echinacoside, poliumoside, acteoside, isoacteosid, 2′-acetylpoliumoside,and 2′-acetylacteoside in Cistanches Herba.Methods The analysis was performed at 20℃, on an Inertsil C18 column(4.6 mm × 250 mm,5.0μm), eluted with a gradient program using acetonitrile(A)-0.1% phosphoric acid aqueous solution(B) as the mobile phase. The flow rate was 1.0 mL·min-1, and the detection wavelength was 330 nm. Results The linear ranges of six components were 0.240~2.400 μg(r= 0.999 3)for echinacoside, 0.300~3.000μg(r=0.9998)for poliumoside, 0.072~0.720μg(r=0.999 4)for acteoside,0.006~0.060μg(r=0.999 1)for isoacteosid,0.150~1.500μg(r=0.999 9)for 2′-acetylpoliumoside, and 0.030-0.300μg(r=0.999 3)for 2′-acetylacteoside. The average recoveries(n=3) of six components were between 98.81%-101.52% with RSD of 0.3%-2.8%. The results showed that the highest contents of echinacoside, acteoside, and isoacteosid were found in Cistanche deserticola and Cistanche tubulosa. Cistanche deserticola contained 2’-acetylacteoside, while Cistanche tubulosa could not be detected. Both of them did not contain poliumoside and 2′-acetylpoliumoside.The main components are echinacoside, acteoside, and 2′-acetylacteoside,and a small amount of poliumoside in Cistanche salsa.The characteristic constituents of Cistanche sinensis are poliumoside and 2′-acetylpoliumoside, and the content of acteoside and 2′-acetylacteoside is very low. Echinacoside could not be detected in it.Conclusion The HPLC method established in this study can distinguish differemt Cistanches Herba, the method is simple, accurate and reproducible, is expected to provide scientific basis for quality evaluation of Cistanches Herba.
