摘 要:目的建立并比较全国不同产地10批鲜马齿苋样品的HPLC指纹图谱,为考察鲜马齿苋的质量及其相关产品开发提供依据。方法采用高效液相色谱法,构建鲜马齿苋指纹图谱,色谱柱为Kromasil C18柱(5μm,250mm×4.6mm),流动相为乙腈甲醇(7∶3)-0.1%甲酸水进行梯度洗脱;进样量20μL,检测波长为320nm,柱温30℃,流量1mL·min-1。使用中药色谱相似度评价系统与SPSS软件分别进行相似度和系统聚类的分析。结果确定了鲜马齿苋指纹图谱中的11个共有峰,峰3咖啡酸为参照峰,相似度评价结果差异较大。聚类分析结果发现廊坊和石家庄归为一类;滨州与红河归为一类;成都和眉山归为一类;洛阳归为一类,其余的可以归为一类。结论不同产地的鲜马齿苋成分、含量差异很大,该方法在一定程度上为今后的鲜马齿苋的应用和研究提供了依据。Objective To establish a HPLC fingerprints analysis method of fresh Portulaca oleracea from 10 batches from different producing areas, and to provide some basis for the quality evaluation of fresh Portulaca oleracea.Methods High performance liquid chromatography(HPLC) was used with Kromasil C18 column(5μm, 250 mm×4.6 mm), the mobile phase was acetonitrile-methanol(7∶3)-0.1% formic acid water for gradient elution, the injection volume was 20 μL, the detection wavelength was 320 nm, the column temperature was 30℃, and the flow rate was 1 mL·min-1. Using chromatographic similarity of traditional Chinese Medicine The evaluation system and SPSS software are used to analyze the similarity and system clustering, respectively.Results The results showed that there were 11 common peaks in the fingerprint of the fresh horse tooth, and the peak of the peak 3 was the reference peak, and the similarity evaluation result was very different. The result of cluster analysis finds that Langfang and Shijiazhuang are classified into one group, Binzhou and Red River are classified into one group, and Chengdu and Meishan are classified as one type;Luoyang is classified into one group, and the rest can be classified as one type. Conclusion The composition and content of the fresh madontia has a large difference due to regional and other reasons, but the method also provides a basis for the application and research of the fresh madontia in the future.
