摘  要:目的探讨柴胡皂苷对宫颈癌细胞凋亡的影响。方法通过细胞增殖实验、细胞周期分析、间质侵袭实验、细胞凋亡分析等方法,研究柴胡不同成分对宫颈癌细胞株HeLa和CaSki增殖的影响,对检测结果进行统计学分析。结果柴胡皂苷对HeLa和CaSki细胞株的增殖有抑制作用。通过对HeLa和CaSki宫颈癌细胞株的细胞周期进程、增殖、侵袭、凋亡及胱天蛋白酶(Caspase)-3和P53蛋白活性的分析表明,柴胡皂苷具有抑制增殖、侵袭,诱导凋亡的作用。研究发现柴胡皂苷可能以剂量和时间依赖的方式诱导宫颈癌细胞的增殖。柴胡皂苷在细胞周期转换的G1~S期具有重要作用,可诱导细胞周期进程、增殖和侵袭的改变。细胞凋亡实验表明,柴胡皂苷能影响宫颈癌细胞株的凋亡。结论柴胡皂苷通过上调Caspase-3和P53蛋白活性而诱导宫颈癌细胞凋亡。Objective To investigate the effect of Saikosaponin on cervical cancer cell apoptosis.Methods The cell proliferation experiment,cell cycle analysis,interstitial invasion experiment and apoptosis analysis were used to determine the effects of different components of Bupleurum chinense DC on the proliferation of cervical cancer cell lines HeLa and CaSki,and the results were statistically analyzed.Results The analysis of cell cycle progression,proliferation,invasion,apoptosis and caspase-3 and P53 protein activities of cervical cancer cell lines HeLa and CaSki showed that Saikosaponin could inhibit proliferation and invasion and induce apoptosis.The study found that saikosaponinmay induce cervical cancer cell proliferation in a dose-and time-dependent manner.Saikosaponinwas significant in the G1-S phase of cell cycle transformation,which induced the change of cell cycle progression,proliferation and invasion.The apoptosis experiment showed that saikosaponin could induce the apoptosis of cervical cancer cell lines.Conclusion Saikosaponinmay induce the apoptosis of cervical cancer cells by up-regulating caspase-3 and P53 protein activities.
