摘  要:目的探讨清热解毒扶正颗粒对慢性阻塞性肺疾病(chronic obstructive pulmonary diseases,COPD)的部分作用机制。方法采用烟熏及气管内注入脂多糖建立COPD大鼠模型,经清热解毒扶正颗粒治疗后,用ELISA检测血清肿瘤坏死因子-α(tumour necrosis factor,TNF-α)、基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)、基质金属蛋白酶抑制因子-1(metallopeptidase inhibitor-1,TIMP-1)水平;用Western Blot检测肺组织磷脂酰肌醇-3激酶(phosphatidylinositol3-hydroxy kinase,PI3K)、蛋白激酶B(protein kinase B,Akt)、核因子-κB-1(nuclear factor-κB-1,NF-κB-1)蛋白表达。结果造模后大鼠血清的TNF-α、TIMP-1、MMP-9均不同程度的升高,与空白对照组比较,TNF-α、MMP-9升高,有显著性差异(P<0.05),TIMP-1升高无显著性差异(P>0.05)。经清热解毒扶正颗粒灌胃治疗后,大鼠血清的TNF-α、TIMP-1、MMP-9均不同程度的降低。与模型组比较,清热解毒扶正颗粒低、中、高剂量组显著降低TNF-α水平(P<0.01);清热解毒扶正颗粒中剂量组显著降低TIMP-1水平(P<0.01),清热解毒扶正颗粒高剂量组显著降低TIMP-1水平(P<0.05),清热解毒扶正颗粒低剂量组降低TIMP-1水平,但差异不显著(P>0.05);清热解毒扶正颗粒低、中、高剂量组降低MMP-9水平不显著(P>0.05)。造模后磷酸化Akt、NF-κB-1、PI3K蛋白表达不同程度地增高,与空白组比较,模型组增高有显著性差异(P<0.05)。经清热解毒扶正颗粒灌胃治疗后,与模型组比较,清热解毒扶正颗粒高剂量组显著降低磷酸化Akt、NF-κB-1、PI3K蛋白表达水平(P<0.05);清热解毒扶正颗粒中剂量组显著降低磷酸化Akt蛋白表达水平(P<0.05);清热解毒扶正颗粒中剂量组、低剂量组对磷酸化NF-κB-1、PI3K蛋白表达作用不显著(P>0.05),清热解毒扶正颗粒低剂量组对磷酸化Akt作用不显著(P>0.05)。结论清热解毒扶正颗粒可降低炎性因子水平,抑制Akt、NF-κB-1、PI3K蛋白表达,进而抑制炎Objective To explore the partial mechanisms of Qingre Jiedu Fuzheng granules on chronic obstructive pulmonary disease(COPD).Methods The COPD rat model was established by smoke inhalation and intratracheal injection of lipopolysaccharide.After treatment with Qingre Jiedu Fuzheng granules,the serum levels of TNF-α,MMP-9 and TIMP-1 in rats were detected by ELISA.The expression levels of phosphatidylinositol-3-kinase(PI3 K),serine-threonine protein kinase(Akt)and transcriptional nuclear factor-κB-1(NF-κB-1)in lung tissues were detected by Western Blot.Results After modeling,the levels of TNF-α,TIMP-1 and MMP-9 in serum were increased to different degrees.Compared with the blank control group,the increases of TNF-αand MMP-9 were significantly different(P<0.05),while the increase of TIMP-1 was not significantly different(P>0.05).After gavage treatment with Qingre Jiedu Fuzheng granules,the serum levels of TNF-α,TIMP-1 and MMP-9 in rats were decreased to different degrees.Compared with the model group,the serum levels of TNF-αwere significantly decreased in the low,medium and high dose groups of Qingre Jiedu Fuzheng granules(P<0.01).The level of TIMP-1 was significantly reduced in the middle dose group of Qingre Jiedu Fuzheng granules(P<0.01),the level of TIMP-1 was significantly reduced in the high dose group of Qingre Jiedu Fuzheng granules(P<0.05),while the level of TIMP-1 was not significantly reduced in the low dose group of Qingre Jiedu Fuzheng granules(P>0.05).The levels of MMP-9 in the low,medium and high dose groups of Qingre Jiedu Fuzheng granules were reduced and there were not significant different(P>0.05).After modeling,the expression levels of phosphorylated Akt,NF-κB-1 and PI3 K were increased to different degrees.And compared with the blank control group,the increases in the model group were significantly different(P<0.05).After gavage treatment with Qingre Jiedu Fuzheng granules,compared with the model group,the expression levels of phosphorylated Akt,NF-κB-1 and PI3 K in the high dose group of
