摘 要:目的研究褪黑素(RAGE)对糖尿病大鼠心肌组织中丙二醛(MDA)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)含量的影响及糖基化终末产物(AGE)和糖基化终末产物受体(RAGE)表达水平的改变。方法清洁级雄性Wistar大鼠,高脂高糖饮食,配合腹腔注射链脲佐菌素诱导糖尿病模型,随后给予低剂量[10 mg/(kg体重·d)]和高剂量[20 mg/(kg体重·d)]MT灌胃16 w。比色法测定心肌组织中MDA、CAT和GSH-Px含量变化;HE染色观察心肌组织病理改变;超敏二步免疫组化染色测定心肌组织中AGE和RAGE蛋白表达。结果经过MT处理16 w后,与模型组比较,低剂量MT组和高剂量MT组血糖值无明显变化(P>0.05);与模型组比较,低剂量MT组和高剂量MT组心肌组织中MDA含量明显降低,CAT和GSH-Px含量明显升高(P<0.05);HE染色显示低剂量MT组和高剂量MT组心肌结构损伤程度均较模型组轻;与模型组比较,低剂量MT组和高剂量MT组心肌组织中AGE和RAGE蛋白表达明显降低(P<0.05)。结论MT可以降低糖尿病大鼠心肌组织MDA含量,提高CAT和GSH-Px含量;可改善心肌结构变化;降低心肌中AGE和RAGE蛋白的表达。Objective To study the effects of melatonin(MT) on the content of MDA, CAT and GSH-PX, and the expressions of AGE and RAGE in the myocardial tissues of diabetic rats.Methods Eighty clean male Wistar rats were selected, each weighed 180~200 g. After one week’s adaptive breeding, these rats were randomly divided into normal, model, low-and high-dose MT groups. The normal group was fed with basal diets. The model, low-and high-dose MT groups were given high-fat and high-sugar diets for four weeks, Streptozotocin(1%STZ solution 50 mg/kg) was administrated through intraperitoneal injections and then the tail vein blood was collected 72 hours later. The rats with fasting blood-glucose greater than or equal to 16.7 mmol/L were taken as the indication of successful model. Then they were given low(10 mg/kg weight per day) and high dose(20 mg/kg body weight per day) of MT intragastric administration for 16 weeks while the normal group received the same amount of saline. Changes in MDA, CAT and GSH-PX in myocardial tissue of rats were determined by colorimetry;pathological changes of myocardium in rats were observed by HE staining;the expressions of AGE and RAGE in myocardium of rats were determined by two step immunohistochemical staining.Results After MT treatment for 16 weeks, compared with the model group, blood glucose value in the low-dose and high-dose MT groups rats had no significant change(P>0.05);however the content of MDA in the myocardial tissue of low-and high-dose MT groups were significantly lower(P<0.05), and the content of CAT and GSH-PX were significantly higher(P<0.05). Compared with the model group in HE staining, myocardial injury degree in low-and high-dose MT groups were lighter whereas the positive expressions of AGE and RAGE in low-and high-dose MT group were significantly lower(P<0.05).Conclusions MT has no significant effect on blood glucose levels in diabetic rats. However it could reduce the content of MDA in myocardial tissue of diabetic rats and increase the content of CAT and GSH-PX MT cou
