摘 要:通过测定彭波半细毛藏系母绵羊AMH激素含量,判定不同年龄阶段母羊的卵巢储备功能,确定无繁殖能力母羊的年龄上限,提高藏系绵羊的养殖效率。试验通过采集0.5~9.5岁不同年龄彭波半细毛藏系母绵羊颈静血每组各5只,1500 r/min离心15 min后取血清,采用酶联免疫吸附(ELISA)法测定0.5~9.5岁彭波半细毛藏系母绵羊50个样本中的AMH血清含量。结果表明,彭波半细毛藏系母羊在0.5~1.5岁阶段AMH含量相对最高,分别为7.6774±3.1806 pg/mL和8.0673±2.8938 pg/mL;2.5~5.5岁AMH含量从5.5628±2.2902 pg/mL下降到5.4551±0.9973 pg/mL,呈逐渐下降趋势;6.5岁开始下降急剧直至9.5岁1.0402±0.5817 pg/mL。此外,4.5岁、6.5岁、8.5岁和9.5岁分别与0.5~1.5岁差异极显著(P<0.01);6.5岁与2.5岁、5.5岁,7.5岁与0.5~1.5岁,8.5岁与2.5岁,9.5岁与5.5岁都差异显著(P<0.05);6.5岁之后各年龄段均不显著,且从6.5岁开始AMH浓度含量低于3.0000 pg/mL以下。结果表明,彭波半细毛藏系母绵羊AMH含量随年龄的增高而降低,呈负相关,说明卵巢储备功能随之减弱,从6.5岁开始卵巢储备功能降低至无繁殖能力,可界定为淘汰年龄上限。An upper age limit of non-breeding ewes and ovarian reserve function were predicted by measuring the content of AMH in serum of Pengbo semi-fine wool Tibetan ewes to raise breeding efficiency.In the experiment,jugular?venous?blood of five sheep from PengBo semi-fine wool Tibetan ewes aged from 0.5 to 9.5 years old were collected from each group.The serum were extracted after centrifuging at 1500 r/min for 15 min,and the content of AMH in 50 samples were determined by the method of ELISA.The results showed that the content of AMH was the highest from 0.5 to 1.5 years old in Pengbo ewes,which was 7.6774±3.1806 pg/mL and 8.0673±2.8938 pg/mL,respectively.From 2.5 to 5.5 years old,the content of AMH reduced gradually with ages from 5.5628±2.2902 pg/mL to 5.4551±0.9973 pg/mL.It began to decrease rapidly from 6.5 years old to 9.5 years old and reached 1.0402±0.5817 pg/mL.In addition,there were very significant(P<0.01)between 4.5,6.5,8.5 and 9.5 years old with ages range from 0.5 to 1.5,respectively.There were significant differences(P<0.05)between 6.5 with 2.5 and 5.5 years old,7.5 with ages range from 0.5 to 1.5,8.5 with 2.5 years old,and 9.5 with 5.5 years old.There was no significant difference all the ages after 6.5,and AMH content were lower than 3.0000 pg/mL.The results showed that the AMH content of PengBo semi-fine wool Tibetan ewes decreased with the increase of age,showing a negative correlation,indicating that the ovarian reserve function decreased with the increase of age.From the age of 6.5,the ovarian reserve function decreased to no reproductive capacity,which can be defined as the upper limit of elimination age.
