摘 要:研究虹鳟(Onco rhynchus mykiss)生长激素基因(GH)的遗传特征和变异分化,采用PCR扩增、电泳及测序等方法,对道氏虹鳟和日本金鳟共33个个体的生长激素基因内含子核苷酸序列进行分析。结果扩增得到的目的基因片段长度为267 bp,两种鱼GH的T、C、A、G四种核苷酸的平均含量分别为30.9%、15.2%、32.0%和21.9%;A+T含量为62.9%,G+C含量为37.1%,A+T含量明显高于G+C含量。后对两种鱼GH单核苷酸多态性(SNP)和单倍型进行分析,共检测到11个多态位点,约占所测核苷酸总长的4.12%。由此界定了14个单倍型,其中日本金鳟单倍型为7个,其单倍型多样度和核苷酸多样度分别为0.74242和0.00799,平均核苷酸差异数K为2.03030;道氏虹鳟群体单倍型也为7个,其单倍型多样度和核苷酸多样度分别为0.91905和0.01012,平均核苷酸差异数K为2.57143。由此可见道氏虹鳟遗传多样性较日本金鳟高,此结果可为以后鲑鳟鱼的分子选择育种及种质资源保护奠定良好的基础。In order to study the genetic characteristics and variation differentiation of growth hormone gene(GH)in Onco rhynchus mykiss,PCR amplification,electrophoresis and sequencing were used to analyze the nucleotide sequence of growth hormone gene introns in 33 individuals of Donaldson rainbow trout and Albino rainbow trout.The results showed that the length of the target gene fragment was 267 bp,and the average contents of T,C,A and G nucleotides in GH were 30.9%,15.2%,32.0%and 21.9%,the content of A+T was 62.9%,the content of G+C was 37.1%,and the content of A+T was significantly higher than that of G+C.After analysis of Donaldson rainbow trout and Albino rainbow trout GH single nucleotide polymorphisms(SNP)and haplotypes,a total of 11 polymorphic sites were detected,accounting for about 4.12%of the total length of the tested nucleotides.Thus,14 haplotypes were defined,including 7 haplotypes in Albino rainbow trout,and haplotype diversity and nucleotide diversity were 0.74242 and 0.00799.The average number of nucleotide differences K was 2.03030.There are also 7 haplotypes in the Donaldson rainbow trout.The haplotype diversity and nucleotide diversity are 0.91905 and 0.01012,respectively,and the average number of nucleotide differences K is 2.57143.It can be seen that the genetic diversity of Donaldson rainbow trout is higher than that of Albino rainbow trout.This result can lay a good foundation for the molecular selection and breeding of salmon trout and the protection of germplasm resources in the future.
