摘 要:对大肠杆菌进行化学诱变获得赖氨酸缺陷型菌株(Lys菌株),在此基础上进一步诱变并筛选出甲硫氨酸缺陷型菌株(Met菌株),最终获得双重缺陷型菌株(Met-Lys菌株),从而提高L-苏氨酸的产量;通过L-苏氨酸和四氯苯醌的荷移反应,可测定普通大肠杆菌、Lys型大肠杆菌和Met-Lys型大肠杆菌发酵后发酵液中L-苏氨酸的含量。结果表明:Lys型大肠杆菌的L-苏氨酸产量比普通型大肠杆菌增加了26.01%,达到了4.07 g/L;而Met-Lys型大肠杆菌的L-苏氨酸产量比普通大肠杆菌增加了299.38%,达到了12.90 g/L。Through chemical mutagenesis of Escherichia coli,the Lysine defective strain(Lys strain)was obtained after screening.On this basis,further mutagenesis and screening of methionine defective strain(Met strain)were carried out,and finally the double defective strain(Met-Lys strain)was obtained,so as to improve the yield of L-threonine.Through the charge transfer reaction of L-threonine and tetrachlorobenzoquinone,the content of L-threonine in the fermentation broth of common Escherichia coli,Lysine-deficient Escherichia coli,Lysine and Methionine-deficient Escherichia coli could be determined.The results showed that the L-threonine production of Lysine-deficient Escherichia coli increased by 26.01% to 4.07 g/L compared with that of normal Escherichia coli.The L-threonine production of Lysine and Methionine double defective Escherichia coli increased by 299.38% compared with that of ordinary Escherichia coli,reaching 12.90 g/L.
