摘  要:目的观察脓毒症小鼠脾脏CD4+T细胞的miR-142-3p的表达水平。方法通过盲肠结扎穿孔法建立脓毒症小鼠模型,28只C57BL/6小鼠随机(随机数字法)分为两组:假手术组,脓毒症模型组,每组各14只。所有小鼠均于手术后48 h处死并取材。采用酶联免疫吸附法检测小鼠血清IL-1β、TNF-α、IL-6、IL-2,通过实时荧光定量PCR(qRT-PCR)方法检测小鼠脾脏CD4+T细胞miR-142-3p的水平。结果脓毒症小鼠脾脏CD4+T细胞的miR-142-3p表达水平较假手术组显著升高(P<0.05),IL-1β、TNF-α、IL-6、IL-2水平均较假手术组显著升高(P<0.05)。结论脓毒症小鼠脾脏CD4+T细胞中miR-142-3p表达上调与炎症相关。Objective To observe the expression level of miR-142-3p in CD4+T cells in the spleen of mice with sepsis.Methods Mice with sepsis was induced by means of cecal ligation and puncture(CLP),28 mice on a C57BL/6 background were randomly divided into two groups:14 in the sham surgery group and 14 in the sepsis model group.All mice were sacrificed at 24 h post-surgery and samples were collected.The serum samples were used to measure the level of IL-1β、TNF-α、IL-6、IL-2 by ELISA,and the expression level of miR-142-3p in CD4+T cell in the spleen was measured by qRT-PCR(Quantitative real time polymerase chain reaction).Results In the sepsis model group,the expression level of miR-142-3p in CD4+T cell in the spleen was increased dramatically compared to those of the sham surgery group(P<0.05),as well as the level of IL-1β、TNF-α、IL-6、IL-2 in the serum samples(P<0.05).Conclusion miR-142-3p in CD4+T cell in the spleen of mice with early sepsis were up-regulated.
