摘  要:优化荧光分光光度法测定瑞舒伐他汀钙含量的方法,建立方法学,并用紫外-分光光度法验证结果的可靠性。在激发波长250nm,发射波长516nm的条件下采用荧光分光光度法测定瑞舒伐他汀钙片中的瑞舒伐他汀钙的含量。以荧光强度为指标,考察pH值、甲醇浓度和时间等3个因素,选定最佳条件,结果的准确性和可靠性用紫外-分光光度法进行验证。荧光强度与瑞舒伐他汀钙浓度在3.17~39.72mg·L^-1范围内具有良好的线性关系,回归方程F=13.96 C+145.71(r=0.9998),检出限为0.38mg·L^-1,平均回收率为98.4%,RSD 1.05%。最佳实验条件为:pH值6.7,时间10 min,甲醇浓度为95%。对正交试验数据进行统计分析,具有统计学意义(P<0.05)。结果显示用荧光分光光度法测定瑞舒伐他汀钙含量特异性好,灵敏度高,检测限低。采用紫外-分光光度法进行验证,结果一致。Tooptimum andestablish a method for determination of rosuvastatin calcium by fluorescence spectrophotometry and compared with UV spectrophotometry.Determination of rosuvastatin calcium in rosuvastatin calcium tablets under the condition of 250nm excitation wavelength and 516nm emission wavelength by fluorescence spectrophotometry.Examine pH,methanol concentration and time increasing to select the best conditions.The fluorescence intensity has a good linear relationship with the rosuvastatin calcium concentration in the range of 3.17~39.72mg·L^-1,regression equation:F=13.96 C+145.71(r=0.9998),detection limit:0.38mg·L^-1,average recovery rate:98.4%(RSD=1.05%).Under the condition of pH=6.7,time=10 min,and the methanol concentration=95%,the fluorescence intensity is the highest.Statistical analysis of orthogonal test data showed statistically significant differences(P<0.05).Conclusion:The results showed that the determination of rosuvastatin calcium had good specificity,sensitivity,and low detection limit.The determination of rosuvastatin calcium by fluorescence spectrophotometer or UV spectrophotometry,lead to consistent results.
