摘 要:筛选一株具有吸附重金属锰能力的菌株,且对其吸附机理和吸附效率进行研究分析。通过16S rDNA序列进行鉴定;用最小抑菌浓度(MIC)测试对不同重金属的耐受程度;研究不同环境因素及锰浓度对菌生长的影响;用电感耦合等离子体发射光谱仪(ICP-AES)、扫描电镜-能谱仪(SEM-EDX)、傅里叶红外显微系统(FT-IR)等仪器对该菌株吸附重金属效率及相关吸附机理进行研究分析。结果发现,16S rDNA序列表明该细菌属于Ralstonia pickettii菌属,命名为ADZH5101。R.pickettii ADZH5101对不同重金属耐受性为Mn2+>Cd2+>Ni2+>Zn2+>Cu2+>Co2+,其中耐受锰浓度可达2200 mg·L^-1。SEM-EDX结果显示吸附后,细胞表面有晶体析出,锰离子含量占总元素16.19%;FT-IR结果显示参与吸附过程的官能团包括—PO43-、胺基中的—C—N—、—M—O(O—M—O)、酰胺基(—CO—NH—)和C—H基团。同时ICP-AES结果显示在锰浓度为450 mg·L^-1时去除效果最好,达到180 mg·L^-1左右。A strain able to adsorb heavy metal manganese was screened,isolated and identified.The strain’s tolerance to several heavy metals was evaluated based on minimal inhibitory concentration(MIC).And the influence of different environmental factors and manganese concentration on the growth was investigated.The adsorption efficiency and related adsorption mechanism of heavy metals were studied by inductively coupled plasma emission spectrometer(ICP-AES),scanning electron microscope-energy spectrometer(SEM-EDX),and fourier transform infrared spectrometer(FT-IR).The isolated strain belonged to Ralstonia pickettii based on its 16S rDNA sequence and was designated as ADZH5101.The tolerance of R.pickettii ADZH5101 to several heavy metals was Mn^2+>Cd^2+>Ni^2+>Zn^2+>Cu^2+>Co^2+.The R.pickettii ADZH5101 can grow in the presence of Mn2+up to 2200 mg·L^-1.The results of SEM-EDX showed that crystals were precipitated on the surface of the cells after adsorption,and the content of manganese ions accounted for 16.19%of the total elements.The FT-IR results showed that the functional groups involved in the adsorption process included-PO43-,-C-N-in the amine group,M—O(O—M—O),amide(—CO—NH—)and C—H groups.The results of ICP-AES showed that the highest removal efficiency of Mn2+by R.pickettii ADZH5101 reached to^180 mg·L^-1 when the concentration of dosed manganese in the medium was 450 mg·L^-1.
