摘  要:【目的】为探讨对节白蜡自然资源的濒危机制,为其野生资源的保护提供依据。【方法】利用cpSSR分子标记,对我国特有的珍稀濒危保护植物对节白蜡10个自然居群196个个体进行遗传多样性研究,并构建其UPGMA聚类图。【结果】6对cpSSR引物扩增出了30条多态性带,居群的遗传多样性(多态性位点百分率:PPL=25.75%,Nei’s基因多样性指数:h=0.0974,香农信息指数:I=0.1440,等位基因数:Na=1.2575,有效等位基因数:Ne=1.1712)低于其在物种水平上的多样性(PPL=75.00%,h=0.1720,I=0.22732,Na=1.7500,Ne=1.2756)。居群间遗传分化系数(Gst)为0.4187,基因流(Nm)为0.6942,遗传距离范围0.0329~0.1569,根据遗传一致度(0.8548~0.9676),10个自然居群聚成4个大类。【结论】对节白蜡总遗传多样性处于中等水平,遗传变异主要发生在居群内(58.11%),因此,保护对节白蜡天然种群应重点保护遗传多样性丰富的居群。【Objective】The present paper aimed to explore the endangered mechanism of natural resources and provide basis for the conservation of wild Fraxinus hupehensis.【Method】The genetic diversity of 196 individuals from 10 natural populations of Fraxinus hupehensis were analyzed by using cpSSR molecular markers,and the the UPGMA cluster of 10 populations was constructed.【Results】The 30 polymorphic bands were amplified from 6 primers.The genetic diversity at populations level(Percentage of polymorphic loci:PPL=25.75%,Nei’s gene diversity index:h=0.0974,Shannon’s information index:I=0.1440,the observed number of alleles:Na=1.2575,the effective number of alleles:Ne=1.1712)was lower than that at the species level(PPL=75.00%,h=0.1720,I=0.22732,Na=1.7500,Ne=1.2756).The Nm(gene flow)value and the cofficient of gene differentiation(Gst)was 0.6942 and 0.4189,respectively,and the genetic distance ranged from 0.0329 to 0.1569.Based on the genetic identity(0.8548-0.9676),10 natural populations were divided into 4 groups.【Conclusion】The total genetic diversity of F.hupehensis was at a moderate level.The genetic variation mainly occurred within the populations(58.11%),and the genetic differentiation among the populations was not obvious.Therefore,it is important to protect the populations with rich genetic diversity.
