针药结合治疗缺血性卒中后失眠大鼠NE、5-HT的实验研究

摘　　要：目的观察柴胡理中汤结合针刺对缺血性卒中后失眠大鼠血清中NE、5-HT含量的影响,探讨柴胡理中汤和针刺治疗失眠症作用机制。方法将40只雄性SD大鼠随机分为空白组、模型组、针刺组、中药组、针药组5组,每组8只。空白组大鼠腹腔注射0.9%氯化钠溶液,每日1次,连续6 d。模型组、针刺组,中药组、针药组在造模成功后分别给予不做任何治疗、针刺、柴胡理中汤、针药结合处理,共6 d。ELISA法检测血清中NE、5-HT含量。结果针药组NE含量显著低于模型组(P <0.05),但与针刺组和中药组无统计学差异(P>0.05),5-HT含量上,针药组显著高于模型组、针刺组和中药组(v<0.05),其数值接近空白组(P>0.05)。结论柴胡理中汤结合针刺可以使卒中后失眠SD大鼠减少NE含量,升高5-HT含量,初步表明柴胡理中汤结合针刺对治疗卒中后失眠症有效。Objective To observe the effect of Chaihu Lizhong decoction combined with acupuncture on the contents of NE and 5-HT in rats with insomnia after ischemic stroke,and to explore the mechanism of Chaihu Lizhong decoction and acupuncture in the treatment of insomnia.Methods 40 male SD rats were randomly divided into 5 groups:blank group,model group,acupuncture group,traditional Chinese medicine group and combination of acupuncture and traditional Chinese medicine group,with 8 rats in each group.Rats in the blank group were intraperitoneally injected with 0.9%sodium chloride solution once a day for 6 consecutive days.The model group,acupuncture group,traditional Chinese medicine group and combination of acupuncture and traditional Chinese medicine group were treated with no treatment,acupuncture,chaihu lizhong decoction and acupuncture and medicine for 6 days after the success of modeling.The contents of NE and 5-HT in serum were determined by ELISA.Results The content of NE in the combination of acupuncture and traditional Chinese medicine group was significantly lower than that in the model group(P<0.05),but there was no statistical difference between the acupuncture group and the traditional Chinese medicine group(P>0.05).The content of 5-HT in the combination of acupuncture and traditional Chinese medicine group was significantly higher than that in the model group,the acupuncture group and the traditional Chinese medicine group(P<0.05),and the value was close to that in the blank group(P>0.05).Conclusion Chaihu Lizhong decoction combined with acupuncture can reduce NE content and increase 5-HT content in SD rats with insomnia after stroke.
