小通草配方颗粒化学成分研究

摘  要:目的对小通草配方颗粒中的化学成分进行研究。方法通过薄层色谱以及高效液相色谱法对小通草化学成分进行初步的分离,然后采用液质联用技术,对所分离开的成分进行定性鉴别,再采用液相制备方法对其主要成分进行制备,运用质谱以及核磁共振方法对制备所得出化学成分进行鉴别,再对鉴别出的主要化学成分进行含量测定。接着用水提取多糖,发生苯酚-浓硫酸显色反应,通过紫外分光光度法测定吸光度并得出小通草颗粒中的多糖含量。结果分离出主要成分为松柏醇。小通草颗粒中多糖含量较高。结论小通草药材以及配方颗粒吸收峰最大成分为松柏醇且松柏醇高效液相方法测定方便简洁,线性关系良好。小通草颗粒中绝大多数成分以多糖形式存在。Objective To study the chemical constituents of Stachyuri Medulla Helwingiae Medulla dispensing granules.Methods The chemical constituents of Stachyuri Medulla Helwingiae Medulla were separated by thin layer chromatography and high performance liquid chromatography,and the separated components were identified qualitatively by liquid mass spectrometry(LC-MS).Then the main components were prepared by liquid phase preparation method,the chemical components were identified by mass spectrometry and nuclear magnetic resonance,and the content of the main components in the formula particles was determined.Then the polysaccharide was extracted with water,and a phenol-sulfuric acid color reaction occurred.The absorbance was determined by ultraviolet spectrophotometry and the polysaccharide content in Xiaotongcao granules was obtained.Results The main component was coniferyl alcohol.The content of polysaccharides in Xiaotongcao granules was higher.Conclusion The maximum absorption peak of Pinus tabulaeformis and formula particles is pine and cypress alcohol and the determination of pine and cypress alcohol by HPLC is convenient and simple,and the linear relationship is good.Most of the ingredients in Stachyuri Medulla Helwingiae Medulla formula granules exist as polysaccharides.
