摘  要:目的:探索加热与未加热毫针对5-盐酸氨酮戊酸体外经皮渗透的影响。方法:通过病理组织切片观察毫火针和毫针在SD幼鼠皮肤中产生的微孔道,检测针刺后不同时间点SD幼鼠皮肤的经皮水分丢失分析皮肤屏障受损情况,再通过经典的垂直透皮扩散实验结合高效液相色谱法HPLC计算5-盐酸氨酮戊酸的体外透皮累积量。结果:HE染色可见毫火针组微孔道深度较未加热的毫针组深(P<0.001)。TEWL数据可以看出,皮肤屏障受损后,毫火针组与毫针组在24小时内具有较快的修复能力。HPLC艾拉累积透皮量结果显示,毫火针和毫针组5-盐酸氨酮戊酸累计透皮量都明显高于对照组(P<0.001),且毫火针比毫针组更具有优势(P<0.001)。结论:毫火针与毫针作用于SD幼鼠皮肤均有利于5-盐酸氨酮戊酸的吸收,毫火针促渗效果更明显。Objective:To explore the effect of heated and unheated needle on percutaneous permeation of 5-ALA in vitro.Methods:The micropores produced by filiform-fire needle and filiform needle in the skin of SD rats were observed by pathological tissue section,and the damage of skin barrier was measured by detecting the transepidermic water loss at different time points.The classical vertical transdermal diffusion experiment combined with high performance liquid chromatography was used to calculate the in vitro transdermal accumulation of 5-ALA after intetvention.Results:HE staining showed that the micropore depth of the filiform-fire needle group was deeper than that of the unheated filiform needle group(P<0.001).The TEWL data showed that the filiform-fire needle group and filiform needle group had a rapid skin barrier repair ability within 24 hours.The results of HPLC showed that the cumulative transdermal volume of 5-ALA in the filiform-fire needle group and the filiform needle group was significantly higher than the controlled group(P<0.001),and the filiform-fire needle group was more advantageous than the filiform needle group(P<0.001).Conclusion:The effect of filiform-fire needle and filiform needle on the skin of young SD rats is beneficial to the absorption of 5-ALA,and the filiform-fire needle has a more obvious effect of promoting penetration.
