摘 要:目的:观察黄芩苷联合TRAIL对人肺癌细胞株A549增殖、凋亡的影响。方法:采用2mg/L、4mg/L、8mg/L黄芩苷和100ng/mLTRAIL单用或联用处理肺癌细胞株A549。利用MTT法检测各组细胞增殖,倒置荧光显微镜观察细胞形态,流式细胞术检测细胞凋亡,RT-PCR法及Western blot法检测Survivin、X连锁凋亡抑制蛋白基因(XIAP)、死亡受体4(DR4)及死亡受体(DR5)mRNA及蛋白相对表达量。结果:相比较于对照组,黄芩苷和TRAIL单用或联用均能呈浓度依赖性抑制A549细胞增殖,诱导其凋亡(P<0.05),而黄芩苷和TRAIL联用比单用时细胞抑制率及凋亡率更高(P<0.05)。相比较于对照组,黄芩苷组和TRAIL组A549细胞数明显降低,漂浮细胞数增多,Survivin及XIAP mRNA及蛋白表达量显著降低,DR5mRNA及蛋白表达量显著升高(P均<0.05),而DR4mRNA及蛋白表达无明显差异。与黄芩苷组或TRAIL组比较,联用组A549细胞数明显降低,漂浮细胞数增多,Survivin及XIAP mRNA及蛋白表达量显著降低,DR5mRNA及蛋白表达量显著升高(P均<0.05),而DR4mRNA及蛋白表达无明显差异。结论:黄芩苷联合TRAIL可协同抑制肺癌细胞株A549增殖,并促进凋亡,其作用机制可能与两者联用能增加死亡受体DR5的表达,进而抑制Survivin和XIAP蛋白表达有关。Objective:To explore the effect of baicalin combined with TRAIL on proliferation and apoptosis of human lung cancer cell line A549.Methods:Lung cancer cell line A549 was treated with 2,4,8 mg/L baicalin and 100ng/ml TRAIL alone or in combination.The proliferation of each group was detected by MTT assay.The morphology of the cells was observed by inverted fluorescence microscope.Apoptosis was detected by flow cytometry.The mRNA and protein relative expression levels of Survivin,X-linked Inhibitor of Apoptosis Protein(XIAP),Death Receptor 4(DR4)and Death Receptor(DR5)were detected by RT-PCR and Western Blot.Results:Compared with the controlled group,baicalin and TRAIL alone or in combination inhibited the proliferation of A549 cells and induced apoptosis in a concentration-dependent manner(P<0.05),and the inhibition rate and apoptotic rate by combination of baicalin and TRAIL were higher than those by alone(P<0.05).Compared with the controlled group,the number of A549 cells in the Baicalin group and the TRAIL group was decreased significantly while the number of floating cells was increased,and the mRNA and protein expression levels of Survivin and XIAP were decreased significantly while the mRNA and protein expression levels of DR5 were increased significantly(all P<0.05),and there were no significant differences in mRNA and protein expression levels of DR4.Compared with the Baicalin group or the TRAIL group,the number of A549 cells in the combination group was significantly decreased while the number of floating cells was increased,and the mRNA and protein expression levels of Survivin and XIAP were decreased significantly while the mRNA and protein expression levels of DR5 were increased significantly(P<0.05),and there were no significant differences in the mRNA and protein expression levels of DR4.Conclusions:Baicalin combined with TRAIL can synergistically inhibit the proliferation of lung cancer cell line A549 and promote apoptosis.Its role mechanism may be related to the increase of DR5 expression and the in
