摘 要:目的:研究复健片对大脑中动脉模闭塞型大鼠cAMP/PKA信号通路mRNA及神经营养因子生长相关蛋白43、脑源性神经营养因子表达的影响,探讨其治疗脑梗死的作用机制。方法:50只SPF级SD大鼠随机分为对照组、模型组、复健片高剂量组、复健片低剂量组、丁苯酞组,每组10只,除对照组外,其余各组制备大脑中动脉闭塞模型,采用Zea Longa 5分制进行评分,1~3分为模型成功。各组采用相应干预措施14d,采用HE染色观察大鼠梗死区神经元损伤情况,RT-PCR法检测梗死区脑组织BDNF、GAP-43、PKA、cAMP mRNA表达情况,Western Blot法检测梗死区脑组织BDNF、GAP-43的含量。结果:与对照组比较,模型组梗死区脑组织神经元损伤明显,GAP-43mRNA表达量降低(P<0.05),BDNF、PKA、cAMP mRNA表达量明显降低(P<0.01),BDNF、GAP-43蛋白表达量降低(P<0.05);与模型组相比,复健片高、低剂量组梗死区脑组织神经元损伤均减轻,BDNF、GAP-43、PKA、cAMP mRNA的表达均明显升高(P<0.01),复健片高剂量组BDNF、GAP-43蛋白表达量明显升高(P<0.01),低剂量组二者的表达也升高(P<0.05)。结论:复健片可激活cAMP/PKA信号通路,调控梗死区脑组织中神经营养因子BDNF、GAP-43的表达,促进大脑中动脉闭塞模型大鼠的神经再生。Objective:To study the effect of Fujian Tablets on mRNA,Growth Associated Protein 43 and Brain-derived Neurotrophic Factor expression in cAMP/PKA signaling pathway for rats with middle cerebral artery occlusion model,and to explore the mechanism of treatment for cerebral infarction.Methods:Fifty SPF SD rats were randomly divided into controlled group,model group,high-dose group of Fujian Tablets,low-dose group of Fujian Tablets,and Butylphthalide group,with 10 rats in each group,except for the controlled group.The middle cerebral artery occlusion model was prepared and scored using the Zea Longa 5-point system,and 1-3 was successfully divided into models.The corresponding interventions were used for 14 days.The neurons in the infarcted area were observed by HE staining.The expressions of BDNF,GAP-43,PKA and cAMP mRNA in the infarcted area were detected by RT-PCR.The infarcted brain was detected by Western Blot.The content of BDNF and GAP-43 was organized.Results:Compared with the controlled group,neuronal damage was observed in the infarcted area of??the model group,GAP-43 mRNA expression decreased(P<0.05),BDNF,PKA,cAMP mRNA expression decreased significantly(P<0.01).The expression of BDNF,GAP-43 protein was decreased(P<0.05).Compared with the model group,the neuronal damage in the infarcted area of??the high and low dose groups was alleviated,and the expression of BDNF,GAP-43,PKA and cAMP mRNA were significantly increased.The expression of BDNF and GAP-43 protein in the highdose group was significantly increased(P<0.01),and the expression in the low-dose group was also increased(P<0.05).Conclusion:Fujian Tablets can activate cAMP/PKA signaling pathway,regulate the expression of neurotrophic factors BDNF and GAP-43 in infarcted brain tissue,and promote nerve regeneration in rats with middle cerebral artery occlusion.
