摘  要:目的:观察何首乌水溶性提取物在体外对血管平滑肌的舒张作用。方法:采用DMT Myograph肌动描计系统记录离体血管张力。观察何首乌水溶性提取物(10-5 mg/ml^10-1 mg/ml)--(二苯乙烯苷含量为65.33%)对基础状态大鼠肠系膜上动脉舒缩的影响;检测内皮完整和去内皮的微动脉血管在缩血管物质高钾和PE的诱导下何首乌水溶性提取物对微动脉的舒张率。结果:何首乌水溶性提取物对基础状态的大鼠肠系膜上动脉血管环(完整内皮)无明显的收缩或舒张作用,且实验前后,血管收缩能力无明显改变,因此推测何首乌水溶性提取物中主要活性成分二苯乙烯苷对血管无直接毒性;何首乌水溶性提取物可以浓度依赖的方式舒张PE或KCl预收缩的大鼠肠系膜上动脉,且何首乌水溶性提取物对PE(10μmol/L)或高钾(60 mmol/L)预收缩的内皮完整大鼠肠系膜上动脉的舒张作用显著强于去除了内皮的大鼠肠系膜上动脉。结论:何首乌水溶性提物取可直接作用于血管,产生明显的舒血管作用,该作用部分依赖血管内皮细胞。其中主要的水溶性成分--二苯乙烯苷可能是具有舒血管作用的有效组分之一。Objective:To observe the relaxation effect of the water-soluble extract from Polygonum multiflorum thunb on vascular smooth muscle in vitro.Methods:DMT myograph system was used to record the vascular tension in vitro.The effect of PME(water-soluble extract from Polygonum multiflorum Thunb,the content of stilbene glycoside was 65.33%)on the relaxation and contraction of the superior mesenteric artery was observed in the basic state rats,and the relaxation rates of the intact and de endotheliated microvasculature induced by high potassium and PE were detect.Results:PME had no relaxation or contraction effect on superior mesenteric artery vascular ring(intact)of basic state rats,the vasoconstriction capacity had no change before and after the treatment,which suggested that stilbene glycoside in PME had no direct vascular toxicity.PME relaxed the superior mesenteric artery induced by PE or KCl,in a concentration dependent manner,and the relaxation effect of PME on the contracted endothelium intact superior mesenteric artery induced by PE(10μmol/L)or high potassium(60 mmol/L)was significantly stronger than that on the endothelium removed superior mesenteric artery of rats.Conclusion:PME could directly affect the blood vessels to produce obvious vasorelaxing effect,which is partly dependent on the vascular endothelial cells.Stilbene glycoside,the main water-soluble component,may be one of the active components with vasoconstriction effect.
