补血益母丸对气血双虚模型小鼠的药效作用研究

摘　　要：目的:探讨补血益母丸对气血双虚模型小鼠的作用。方法:以放血和环磷酰胺并用建立气血双虚小鼠模型,模型建立后,连续给予补血益母丸1.82、7.28 g/kg 10 d进行干预。末次给药后,检测小鼠负重游泳时间和全血乳酸含量;动物处死前,检测小鼠游泳后血尿素氮的含量;采用摘眼球法取血行血常规检测;摘取小鼠胸腺和脾脏并称重,计算脏器指数;部分脾脏在4%多聚甲醛中固定后行HE染色观察脾脏形态;部分脾脏用于Western blot实验检测肿瘤坏死因子α(TNF-α)和白介素2(IL-2)蛋白表达情况;取小鼠右腿骨髓,检测骨髓有核细胞。结果:与正常对照组相比,模型对照组出现活动减少,耳发白,呼吸短促等症状,小鼠负重游泳时间显著缩短(P<0.01),脾指数和胸腺指数均明显降低(P<0.05或P<0.01),且血乳酸和血尿素氮含量明显升高(P<0.05或P<0.01);骨髓有核细胞显著减少(P<0.01),血中红细胞数、白细胞数和血红蛋白含量均显著降低(P<0.01);脾脏结节和淋巴细胞数显著减少(P<0.01),红、白髓界线不明,淋巴结和红髓中有广泛的局灶性纤维化;脾脏中IL-2蛋白表达明显下调(P<0.01),TNF-α蛋白表达明显上调(P<0.05)。与模型对照组相比,乳酸亚铁10 ml/kg组、四物汤12 g/kg组、补血益母丸1.82 g/kg、7.28 g/kg组小鼠负重游泳时间均显著延长(P<0.01),脾指数和胸腺指数均明显升高(P<0.01);血乳酸含量均明显降低(P<0.01),骨髓有核细胞显著增多(P<0.01),血中红细胞数、白细胞数和血红蛋白含量均明显升高(P<0.01);脾脏结节和淋巴细胞数明显增多(P<0.01),红、白髓较清晰;小鼠脾脏中IL-2蛋白表达上调(P<0.05或P<0.01),补血益母丸7.28 g/kg组血尿素氮含量明显降低(P<0.01),小鼠脾脏中TNF-α蛋白表达下调(P<0.05)。结论:补血益母丸能有效地抗气血双虚证候。这可能与其改善骨髓造血功能,提高气血双虚模型小鼠外周血细胞数和提高免疫功能有关。Objective:To investigate the effect of Buxue Yimu Pill in mice with Qixueshuangxu syndrome.Methods:The Qixueshuangxu mice model was established by bloodletting method and cyclophosphamide.After the model was established,the mice were given Buxue Yimu Pill by gavage for 10 days.After the last administration,the weight-bearing swimming time and blood lactic acid content of mice were detected.Before mice were sacrificed,the blood urea nitrogen content after swimming was detected,blood was taken from the eyeball for blood routine examination,the mouse thymus and spleen were was collected and weighed,then the organ index was calculated.A part of mice spleen was fixed in 4%paraformaldehyde,and histopathological changes were observed by HE staining.The other part of spleens were used to detect the protein expressions of TNF-αand IL-2 by western blot.The right leg bone marrow was collected to detect bone marrow nucleated cells.Results:Compared with the control group,symptoms like reducing activity,ear turn white,short of breath were observed in the model group.The weight-bearing swimming time in the model group was significantly shortened(P<0.01),the spleen index and thymus index were significantly reduced(P<0.05,P<0.01),the levels of blood lactic acid and blood urea nitrogen were significantly increased(P<0.05,P<0.01),the number of nucleated cells in the bone marrow was significantly decreased(P<0.01),the number of red blood cells,white blood cells and hemoglobin in the blood were significantly reduced(P<0.01),spleen nodules and lymphocytes were significantly decreased(P<0.01),the boundary between red and white pulp was unclear,and extensive focal fibrosis was observed in lymph nodes and red pulp.The IL-2 protein expression in the spleen was down-regulated(P<0.01),and TNF-αprotein expression was up-regulated(P<0.05).Compared with the model group,the weight-bearing swimming time of mice in Buxue Yimu Pill groups(1.82 g/kg、7.28 g/kg)and 12 g/kg Siwutang group was prolonged(P<0.01),the spleen indexes and thymus indexes
