摘  要:目的研究程序性死亡蛋白1(programmed cell death1,PD-1)在小鼠口腔黏膜鳞状细胞癌微环境中的变化和可能作用。方法利用化学致癌剂4-硝基喹啉-1-氧化物(4-nitroquinoline-1-oxide,4NQO)饮水诱导建立C57小鼠口腔黏膜鳞癌动物模型。对小鼠模型口腔黏膜肿瘤病变部位进行组织病理学检测和分析;通过Real-time PCR和流式细胞术检测免疫检查点PD-1基因和蛋白的表达水平。结果在暴露4NQO 28周后,成功诱导小鼠发生口腔黏膜鳞癌。组织病理学结果显示,小鼠发生的肿瘤为口腔黏膜鳞癌。Real-time PCR和流式细胞术检测结果表明,小鼠舌部黏膜肿瘤组织和脾脏中的CD4^+T细胞,PD-1基因和蛋白表达都明显上调。结论免疫检查点PD-1在小鼠口腔黏膜鳞癌发生中出现明显升高,PD-1与口腔黏膜鳞癌的发展可能相关。Objective To identify the changes and potential roles of immune checkpoint PD-1 in oral squamous cell carcinoma(OSCC)microenvironment.Methods The oral squamous cell carcinoma model of C57 mice was induced and established by potable water containing 4-nitroquinoline-1-oxide(4 NQO).Lesion location of the oral squamous cell carcinoma of the mice model was detected and analyzed by histopathology.Real-time PCR and flow cytometry were used to detect the expression level of PD-1 gene and protein.Results OSCC model was successfully induced by exposing to 4 NQO after 28 weeks.Histopathological evaluation showed that oral mucosal squamous cell carcinoma occurred in mice.Real-time PCR and flow cytometry showed that PD-1 gene and protein expression in CD4^+T cell was significantly up-regulated in tongue mucosal tumor tissues and spleen.Conclusion PD-1 shows significant up-regulation in mice OSCC and is possibly related to the development of OSCC.
