摘　　要：为了研究NLRC5基因在SPF大白猪不同组织中的表达和分布,试验以稀释的标准重组质粒为模板绘制标准曲线,建立实时荧光定量PCR方法,检测SPF大白猪肺脏、颌下淋巴结、腹股沟淋巴结、脾脏、空肠、胃、小肠、直肠、肾脏、心脏、回肠、肝脏、结肠、盲肠、扁桃体、肠系膜淋巴结等16种器官中NLRC5基因mRNA的表达差异和分布,并用Western-blot方法检测16种器官中NLRC5基因编码蛋白表达差异和分布。结果表明:NLRC5基因在SPF大白猪16种器官中均能表达,mRNA和蛋白表达水平大体一致,其中在淋巴结、脾脏和扁桃体等免疫器官中的表达量均较高,在胃和肠道等具有黏膜免疫功能的器官中表达量次之,在心脏和肝脏等非免疫器官中的表达量相对较低。说明试验获得了SPF大白猪不同器官中NLRC5基因的天然表达量,并且在免疫器官和具有黏膜器官中有较高的表达。To study the expression and distribution of NLRC5 gene in different organs of SPF Yorkshire breed, a real-time fluorescent quantitative PCR(qRT-PCR) method was established by drawing a standard curve with diluted standard recombinant plasmid as template. The expression and distribution of NLRC5 gene mRNA and protein in 16 different organs samples(lungs, submandibular lymph nodes, inguinal lymph nodes, spleen, Jejunum, stomach, small intestine, rectum, kidney, heart, Ileum, liver, colon, cecum, tonsil and mesenteric lymph nodes) of SPF Yorkshire pigs by real-time qRT-PCR and Western-blot respectively. The results showed that NLRC5 gene could be expressed in 16 kinds of organs of SPF Yorkshire pigs, and the expression levels for the mRNA and protein were almost the same. The expression of NLRC5 gene washigh in immune organs(lymph nodes, spleen, tonsil), followed by the organs with mucosal immune function such as stomach and intestine, and the lowest expression was observed in vital organs such as heart and liver. Our findings revealed the natural expression of NLRC5 gene in different organs from SPF Yorkshire pigs was obtained, and the expression of NLRC5 gene was highly expressed in the immunological organs as well as in mucosal organs.
