摘  要:褐变是制约木本果树离体培养成功的重要因素,探讨苹果离体再生过程中经常遇到的外植体褐化问题,对苹果高效再生和遗传转化体系建立具有重要意义。本研究以柱状苹果‘润太一号’为试材,研究了遮光时间(0, 7 d, 14 d)、消毒时间(6 min, 7 min, 8 min, 9 min)、茎段长度和防褐化剂对外植体褐化率的影响,并用不同激素配比对不定芽进行诱导。结果表明,外植体的褐化率随遮光时间增加而有所降低,但外植体的生长势变弱,以遮光7 d的材料为宜,褐化率为25%。不同腋芽个数对褐化率的影响差异不显著。褐化率随着消毒时间的增加而增加,最佳的消毒时间为8 min,褐化率为23.33%。不同防褐化剂处理的结果发现以培养基中加入100 mg/L Cef褐化率最低。不定芽诱导的结果发现,不定芽诱导率随诱导天数增加而增加,以诱导50 d效果较好,最适宜的不定芽诱导培养基为MS+4.0 mg/L 6-BA+0.5 mg/L NAA。本研究结果可为柱状苹果高效再生体系提供理论依据。Browning is an important factor that restricts the success of the in vitro culture of the woody fruit tree.Study on the browning problem of explants often encountered in the process of apple regeneration in vitro,it is of great significance for the efficient regeneration of apple and the establishment of genetic transformation system.The columnar apple'Runtai No.1'were selected as materials to assess the effects of different shading time(0,7 d,14 d),different disinfection time(6 min,7 min,8 min,9 min),different stem length and different browning in-hibitors on browning rate of explants.The results showed that the browning rate of explants was decreased with the increase of shading time,while the growth potential of explants was weaker.The material was best for 7 days of shading time,and the browning rate was 25%.The number of axillary buds has no significant difference in the effect of browning rate.The browning rate was increased with the increase of disinfection time,the best disinfection time was 8 minutes and the browning rate was 23.33%.The browning rate was the lowest in murashige and skoog(MS)medium supplemented with 100 mg/L cefotaxime sodium(Cef).When the adventitious buds were in-duced,the induction rate of adventitious buds was increased with the increase of the number of days of induction,and the effect of inducing adventitious buds on 50 days was better,and the most suitable adventitious bud induc-tion was MS+4.0 mg/L 6-benzylaminopurine(6-BA)+0.5 mg/L 1-naphthylacetic acid(NAA).The results can provide a theoretical basis for the efficient regeneration system of columnar apple.
