摘 要:受体材料的生理环境是蓝色花卉转基因育种成功的重要因素之一。为了筛选出适宜进行蓝色转基因工程的仙客来受体品种,本研究以12个不同类型的仙客来花瓣为材料,对其二氢黄酮醇4-还原酶利用二氢杨梅素(dihydromyricetin, DHM)生成飞燕草素苷的能力、总黄酮含量、液泡pH值进行检测,结合灰色关联分析方法对各指标进行综合分析。结果表明:仙客来‘条纹荷花’利用二氢杨梅素生成飞燕草素苷的量最多达39.01μg/g,其花瓣pH也最高为6.82,仙客来‘八重玫瑰’的总黄酮含量最多达2 507.64μg/g;由这3个值组成灰色关联分析的参考品种参数,计算出加权关联度最高的品种为仙客来‘八重玫瑰’,其关联度为0.840 8。本研究从不同类型仙客来品种中筛选出了二氢黄酮醇4-还原酶特异性催化能力强、总黄酮含量和液泡pH值高的品种,可以为蓝色仙客来的转基因育种提供材料参考。In order to screen out the cyclamen receptor varieties suitable for blue transgenic engineering.We used 12 different types of cyclamen petals as materials,and detected the ability of DFR to generate delphinidin by dihydromyricetin(DHM),the total flavonoid content of petals,the pH of the vacuole.The gray correlation analysis method was used to comprehensively analyze each index.The results showed that the contents of delphinidin produced by cyclamen'Strip lotus'using DHM was up to 39.01μg/g,and its petal pH was also up to 6.82.The total flavonoid content of cyclamen'Double rose'was up to 2507.64μg/g.These three values is composed of the reference sequence of gray correlation analysis.And the species with the highest weighted correlation is calculated as the cyclamen'Double rose'with a correlation degree of 0.8408.The varieties with strong DFR specificity,total flavonoid content and high vacuolar pH were screened from different types of cyclamen varieties,which could provide reference for transgenic breeding of blue cyclamen.
