摘  要:目的探讨不同剂量乙烯利不同作用时间下对雄性幼鼠睾丸组织的影响。方法将45日龄健康雌性Sprague-Dawley(SD)大鼠32只,采用随机数字表法随机分为对照组和低、中、高剂量乙烯利组,取同日龄健康雄性SD大鼠32只与之一对一合笼。每日分别对雌鼠进行灌胃,低、中、高剂量组分别用200、400、800 mg/kg乙烯利水溶液灌胃,对照组采用等量9 g/L盐水,仔鼠出生后,停止给母鼠灌胃,改为给雄性仔鼠灌胃。子代雄鼠于出生0日龄、14日龄、28日龄时,分别从各组采用随机数字表法随机选取12只处死。取新鲜睾丸组织行苏木精-伊红(HE)染色,光镜下观察睾丸组织形态变化。采用脱氧核糖核苷酸末端转移酶介导的缺口末端标记(TUNEL)法标记凋亡细胞,利用荧光显微镜检测睾丸生精细胞凋亡指数(AI)。结果1.0日龄时高剂量组较中剂量组、低剂量组和对照组生精小管略增粗,生精细胞排列稍紊乱。高剂量组AI(1.00±0.06)明显高于对照组(0.41±0.03),差异有统计学意义(P<0.01);对照组、低剂量组与中剂量组比较差异无统计学意义(P>0.05)。2.14日龄时低、中、高剂量组较对照组生精小管明显增粗,排列疏松,生精细胞数目稀少,排列紊乱。14日龄时低剂量组(2.13±0.10)、中剂量组(2.18±0.10)、高剂量组(3.90±0.23)AI均明显高于对照组(1.00±0.02),差异有统计学意义(F=2508.36,P<0.01)。中、低剂量组比较AI差异无统计学意义(P>0.05)。3.28日龄时低、中、高剂量组较对照组生精小管显著增粗,排列更疏松,生精细胞数目更少,排列严重紊乱。28日龄时低剂量组(5.52±0.13)、中剂量组(9.44±0.07)、高剂量组(14.56±0.27)AI均明显高于对照组(3.11±0.13),差异有统计学意义(F=10784.69,P<0.01)。结论乙烯利可引起新生大鼠及幼鼠雄性睾丸生精小管增粗,生殖细胞排列紊乱,生精细胞凋亡增加和生精能力下降;且大鼠接触乙烯利时间越长,乙烯利浓Objective To explore the effects of different doses of Ethephon on testes of male pups.Methods Thirty-two 45-day-old healthy female Sprague-Dawley(SD)rats were randomly divided into the control group and the low dose,middle dose and high dose Ethephon groups by the random figure table.The female rats in the low dose,middle dose and high dose Ethephon groups were given 200,400,and 800 mg/kg Ethephon solution,respectively.The control group was treated with 9 g/L saline.After the birth of the offspring,the mother rats were not administrated with any medications,and the male offspring rats were given Ethephon solution instead.Twelve offspring male rats were randomly selected from each group and killed at the age of 0,14 and 28 days after birth.Fresh testicular tissues were stained with hematoxylin eosin(HE),and the morphological changes of testicular tissues were observed under light microscope.The apoptotic cells were labeled by terminal dexynucleotidyl transferase(TdT)mediated dUTP nick end labeling(TUNEL)method and the apoptosis index(AI)of spermatogenic cells was detected by fluorescence microscope.Results(1)Compared with the newborn rats in the middle dose group,low dose group and control group,se-miniferous tubules in the newborn rats of the high dose group were slightly thicker,and seminiferous cells were arranged slightly in disorder.The AI of the newborn rats in high dose group was significantly higher than that in the control group(1.00±0.06 vs.0.41±0.03,P<0.01).The AI of the newborn rats in the middle dose group was not significantly different from that in the control group and the low dose group(P>0.05).(2)The seminiferous tubules of the 14-day-old rats in the low dose,middle dose and high dose Ethephon groups were significantly thicker and arranged more loosely than those in the control group.Compared with the control group,there were very few seminiferous cells,which were arranged disorderly in the low dose,middle dose and high dose Ethephon groups.The AI of the 14-day-old rats in the low dose,middle
