摘 要:目的通过紫外线(UV)照射诱发小鼠皮肤光老化并服用潞党参口服液观察,探讨潞党参口服液对Fas/FasL在正常小鼠皮肤和光老化皮肤中表达的影响。方法将90只健康昆明小鼠随机分为空白组、光老化模型组、潞党参高剂量组、潞党参中剂量组、潞党参低剂量组、VE组,每组15只。除空白组外每组均紫外线照射造模12周,造模成功后给予不同剂量的药物治疗。应用免疫组化法(Immunohistochemistry IHC)检测每组小鼠皮肤组织中Fas和FasL的表达情况。结果空白组小鼠皮肤组织中Fas、FasL含量较少;与空白组比较,光老化模型组皮肤组织中Fas表达升高(P<0.05),具有统计学意义;与光老化模型组相比,潞党参治疗组小鼠皮肤组织中Fas表达降低(P<0.05),有统计学意义,其中潞党参高剂量组的效果最好。与空白组比较,光老化模型组皮肤组织中FasL表达升高(P<0.05),具有统计学意义;与光老化模型组相比,潞党参治疗组小鼠皮肤组织中FasL表达降低(P<0.05),结果具有统计学意义。结论潞党参能够下调小鼠皮肤组织中凋亡因子Fas、FasL的表达,减少细胞凋亡,进而抑制皮肤光老化的发生,可能在皮肤组织中发挥抗光老化抗炎抗衰老的作用。Objective:To observe the photoaging of mice skin by ultraviolet(UV)irradiation andobserve the effect of Ludangshen(Codonopsispilosula in Changzhi)oral liquid on the expression of Fas/FasL in normal mice skin and photoaged skin.Methods:Ninety healthy Kunming mice were randomly divided into blank group,photoaging model group,high,middle and low dose groups of Ludangshen(Codonopsispilosula in Changzhi),and VE group,with 15 rats in each group.Except for the blank group,each group was irradiated with ultraviolet light for 12 weeks,and different doses of drugs were given after successful modeling.Immunohistochemistry(IHC)method was used to detect the expressions of Fas and FasL in the skin tissue of mice in each group.Results:A mouse photoaging model was successfully produced.The contents of Fas and FasL in the skin tissue of the blank group were less.Compared with that of the blank group,the expression of Fas in the skin tissue of the photoaging model group was increased(P<0.05),which was statistically significant.Compared with that of the photoaging model group,the expression of Fas in the skin tissue of Ludangshen(Codonopsispilosula in Changzhi)treatment group was decreased(P<0.05),which was statistically significant.The high dose group of Ludangshen(Codonopsispilosula in Changzhi)was the best.Compared with that of the blank group,the expression of FasL in the skin tissue of the photoaging model group was increased(P<0.05),which was statistically significant.Compared with that of the photoaging model group,the expression of FasL in the skin tissue of the mice treated with Ludangshen(Codonopsispilosula in Changzhi)was decreased(P<0.05),and the results were statistically significant.Conclusion:Ludangshen(Codonopsispilosula in Changzhi)can down-regulate the expressions of apoptotic factors Fas and FasL in mice skin tissue,reduce apoptosis,and then inhibit the occurrence of skin photoaging,which may play a role in anti-aging,anti-inflammatory and anti-aging effects in the skin group.
