摘 要:目的研究不同时间点脾虚对高脂血症大鼠胆固醇逆向转运相关蛋白表达的影响,探讨脾虚膏脂转输障碍的机制及时效性。方法实验分3个时间点(2周,10周,16周),其中2周为脾虚造模时间,第10周即高脂喂养8周,16周为高脂喂养14周,在2周、10周及16周末分别取材。脾虚组采用饮食不节加力竭游泳的复合方法造脾虚模型。正常对照组、脾虚组喂饲基础饲料,高脂组、脾虚高脂组喂饲高脂饲料,自由摄食,饮水不限。在2周末正常对照组和脾虚组各6只取材。在10周和16周末正常对照组、脾虚组、高脂组、脾虚高脂组各6只取材。全自动生化分析仪检测血清TC、TG、LDL-C、HDL-C含量;间苯三酚法检测大鼠D-木糖排泄率;碘-淀粉比色法检测血清淀粉酶活力;HE染色观察肝脏细胞形态变化;ELISA法检测血清PLTP、LCAT、CETP含量。结果脾虚造模2周后,与正常对照组相比,脾虚组大鼠尿D-木糖排泄率及AMS含量明显降低(P<0.01)。第10周末,与正常对照组相比,脾虚组和脾虚高脂组大鼠尿D-木糖排泄率及AMS含量明显降低(P<0.01)。与脾虚组相比,脾虚高脂组大鼠尿D-木糖排泄率及AMS含量有进一步下降趋势。第16周末,与正常对照组相比,脾虚高脂组大鼠尿D-木糖排泄率及AMS含量明显降低(P<0.01)。第10周末,正常对照组肝细胞核圆、大而居中,细胞索条理清晰,肝窦正常,与正常对照组相比,高脂组和脾虚高脂组大鼠血清TC、TG、LDL-C、PLTP、CETP含量明显升高,HDL-C、LCAT明显降低(P<0.05或P<0.01),肝细胞肿大变圆,脂肪空泡化明显,肝细胞形态异常,肝小叶结构破坏,肝索排列紊乱,胞浆疏松,部分细胞核挤向胞膜,与高脂组相比,脾虚高脂组大鼠血清TC、LDL-C、PLTP、CETP含量明显升高(P<0.05或P<0.01),TG有升高趋势,LCAT含量明显降低(P<0.05),HDL-C有下降趋势,大鼠肝脏组织泡沫化更为严重;第16周末,与正常对照组相比,高脂组和脾虚高脂组Objective To study the effects of spleen deficiency at different time points on the expressions of cholesterol reverse transport related proteins in hyperlipidemia ratsand explore the mechanism and timeliness of spleen deficiency fat transfer disorder.Methods The experiment was divided into 3 time points(2 weeks,10 weeks,16 weeks),of which 2 weeks were spleen-deficiency modeling time,10 weeks were high-fat feeding for 8 weeks,and 16 weeks were high-fat feeding for 14 weeks.Samples were taken at 2,10 and 16 weeks respectively.In the spleen-deficiency group,a spleen-deficiency model was established by a combination of diet and exhaustive swimming.The normal control group and the spleen deficiency group were fed with basal food,and the high-fat group and the spleen-deficiency high-fat group were fed with high-fat diet and the rats can free feeding and unlimitedly drink.At the end of the 2 weeks,6 rats in normal control group and 6 in spleen deficiency group were sampled.At the end of 10 and 16 weeks,6 rats in the normal control group,spleen deficiency group,high fat group and spleen deficiency high-fat group were taken.The serum levels of TC,TG,LDL-C and HDL-C were detected by automatic biochemical analyzer.The D-xylose excretion rate was detected by phloroglucinol method.The serum amylase activity was detected by iodine-starch colorimetry.HE staining observation morphological changes of liver cells,serum levels of PLTP,LCAT and CETPwere detected by ELISA.Results After 2 weeks of spleen deficiency modeling,the urinary D-xylose excretion rate and AMS content in the spleen-deficiency group were significantly lower than those in the normal control group(P<0.01).At the end of the 10 th week,compared with those of the normal control group,the urinary D-xylose excretion rate and AMS content in the spleendeficiency group and the spleendeficiency high-fat group were significantly lower(P<0.01).Compared with the spleen deficiency group,the urinary D-xylose excretion rate and AMS content in the spleendeficiency high-fat group
