摘  要:目的探讨利用大鼠骨髓间充质干细胞(BMSCs)体外构建成骨细胞膜片并用于修复大鼠颅骨缺损的可行性。方法将大鼠BMSCs以高密度接种于培养皿中,并在体外向成骨细胞诱导3周,待形成膜片后用细胞刮刮起膜片,实验组将成骨细胞膜片植入大鼠8 mm直径颅骨缺损区,在非成骨诱导条件下诱导形成的细胞膜片植入大鼠8 mm直径颅骨缺损区为对照组,同时设置空白组。术后6、12周每组分别取材,进行大体观察、X线检测和组织学检查。结果在体外成功构建出成骨细胞膜片,将膜片植入大鼠颅骨缺损处12周后观察到实验组较对照组有更多的新骨形成,组织学检查证实为新骨。结论本实验成功构建成骨细胞膜片,为人类骨缺损的修复提供了新的方法。Objective To investigate the feasibility of using bone marrow stem cells(BMSCs)in vitro to construct osteoblasts cell sheet and repair skull defect in rats.Methods Rat BMSCs were inoculated in a culture dish with high-density and induced to osteoblasts for 3 weeks.The cell sheet was scraped by cells scrap after the formation of the cell sheet,and the osteoblastic cell sheet was implanted into the 8 mm diameter defect of the rat skull in the experimental group.The cell sheet formed under non-osteogenic induction was implanted into the 8 mm diameter defect area of the rat skull as the control group,and set the blank group.General observation,X-ray examination and histological examination were conducted at 6 and 12 weeks after the operation.Results The osteoblastic cell sheet was successfully constructed in vitro,and more new bone was formed in the experimental group than in the control group at 12 weeks after the cell sheet was implanted in the skull defect of the rats,which was confirmed as new bone by histological examination.Conclusion This experiment successfully constructed osteoblast membranes,which provided a new method for repairing human bone defects.
