摘  要:目的观察宣肺逐饮方对肺癌恶性胸腔积液模型小鼠胸膜粘连的影响及对肺癌恶性胸腔积液的部分作用机制。方法选取C57BL/6小鼠50只,随机分为对照组、模型组、顺铂组和宣肺逐饮方低、高剂量组,每组10只。除对照组外,其余4组均胸膜注射Lewis肺癌细胞,构建肺癌恶性胸腔积液模型。Lewis肺癌细胞注射后第14天,通过计算机断层扫描观察各组小鼠胸腔积液情况,断头处死小鼠后测量胸腔积液体积,观察其右肺胸膜间皮细胞和结缔组织形态。观察并比较各组小鼠细胞外信号调节激酶(ERK)1/2、水通道蛋白4(AQP4)的表达水平。结果与对照组比较,模型组小鼠胸腔积液量、肺组织ERK1/2和AQP4 mRNA、ERK1/2和AQP4相对蛋白表达水平显著增多(P<0.05)。与模型组比较,顺铂组和宣肺逐饮方低、高剂量组胸腔积液量、肺组织ERK1/2、AQP4 mRNA和ERK1/2、AQP4相对蛋白表达水平显著减少,且宣肺逐饮方低剂量组上述指标高于顺铂组(P<0.05)。宣肺逐饮方高剂量组胸腔积液量、肺组织ERK1/2、AQP4 mRNA和ERK1/2、AQP4相对蛋白表达水平与顺铂组比较差异无统计学意义(P>0.05)。结论宣肺逐饮方可促进肺癌恶性胸腔积液模型小鼠胸腔积液的吸收,改善其胸膜粘连,可能与抑制ERK1/2信号通路的激活,降低AQP4蛋白表达水平有关。Objective To investigate the effects of Xuanfei Zhuyin Decoction on pleural adhesion in malignant pleural effusion model mice with lung cancer,and the partial mechanism of Xuanfei Zhuyin Decoction on malignant pleural effusion in mice with lung cancer.Methods Fifty C57BL/6 mice were randomly divided into control group,model group,cisplatin group,low-and high-dose Xuanfei Zhuyin Decoction groups,with 10 mice in each group.Except the control group,Lewis lung cancer cells were injected into the pleura of the other four groups to construct the malignant pleural effusion model of lung cancer.At 14 days after injection of lewis lung cancer cells,the pleural effusion of mice in each group was observed by CT,the volume of effusion was measured after decapitation of mice,and the morphology of mesothelial cells and connective tissue of the right lung was observed.The expression levels of ERK 1/2 and AQP4 were observed and compared.Results Compared with the control group,the volume of pleural effusion,the expression levels of ERK1/2 and AQP4 mRNA,and relative protein expression levels of ERK1/2 and AQP4 in the lung tissue of the model group were significantly increased(P<0.05).Compared with the model group,the above indicators in the cisplatin group and the low-dose and high-dose Xuanfei Zhuyin Decoction groups were significantly reduced,and the above indexes in the low-dose Xuanfei Zhuyin Decoction group were higher than those in the cisplatin group(P<0.05).The volume of pleural effusion,ERK1/2 and AQP4 mRNA as well as their relative protein expression levels in the high-dose Xuanfei Zhuyin Decoction group were not significantly different from those in the cisplatin group(P>0.05).Conclusion Xuanfei Zhuyin Decoction can promote the absorption of pleural effusion and improve pleural adhesion in MPE model mice with lung cancer,which may be related to inhibiting the activation of ERK1/2 signaling pathway and reducing the expression of AQP4 protein.
