摘 要:目的探讨冬凌草甲素对人脑胶质瘤U251细胞放疗敏感性的影响。方法人脑胶质瘤U251细胞随机分为对照组(脑胶质瘤U251细胞常规培养)、冬凌草甲素组(冬凌草甲素10μmol/L培养24 h)、X线照射组(21EX型双光子高能直线加速器照射)、联合放疗组(X线照射+冬凌草甲素)。观察4组人脑胶质瘤U251细胞的增殖抑制率,检测其凋亡率,分析Cleaved Caspase-3、Cleaved Caspase-9蛋白表达水平变化。结果与对照组比较,冬凌草甲素组、X线照射组及联合放疗组人脑胶质瘤U251细胞增殖率降低,且联合放疗组低于冬凌草甲素组及X线照射组(P<0.05);与对照组比较,冬凌草甲素组、X线照射组及联合放疗组人脑胶质瘤U251细胞凋亡率及凋亡相关蛋白的表达量均显著增高,且联合放疗组高于冬凌草甲素组及X线照射组(P<0.05)。结论冬凌草甲素可显著提高人脑胶质瘤U251细胞的放疗敏感性,为冬凌草甲素应用于胶质瘤的放疗提供了理论依据。Objective To investigate the effect of oridoninon on radiosensitivity of human glioma U251 cells.Methods Glioma U251 cells were randomly divided into control group(cells were regular cultured),oridonin group(cells were cultured with 10μmol/L oridonin for 24 h),X-ray irradiation group(21EX two-photon high energy linear accelerator irradiation),and combined radiotherapy group(X-ray irradiation+oridonin).The proliferation inhibition rate of U251 cells was observed,The apoptotic rate were detected,and the expression level of Cleaved Caspase-3 and Cleaved Caspase-9 protein was analyzed.Results Compared with the control group,the cell proliferation rate was significantly reduced in oridonin group,X-ray irradiation group and combined radiotherapy group,which was lower in combined radiotherapy group than in oridonin group and X-ray irradiation group(P<0.05).In addition,apoptosis rate and the expression of apoptosis-related proteins were significantly increased in oridonin group,X-ray irradiation group and combined radiotherapy group,which were higher in combined radiotherapy group than in oridonin group and X-ray irradiation group(P<0.05).Conclusion Oridonin could obviously increase the radiosensitivity in human glioma U251 cells,which provides a theoretical basis for the application of oridonin in radiation therapy of glioma.
