摘 要:目的研究两组不同波长630 nm和810 nm窄带LED照射对体外高糖环境培养下的小鼠成纤维细胞增殖能力的影响。方法实验对象为小鼠成纤维细胞系(L929),以高糖DMEM培养基培养,每孔3 000个细胞接种于96孔板。根据不同波长分为两组分为630 nm LED组和810 nm LED组,每组再根据不同照射时间(0,100,200,500,1 000和2 000 s)分为6个小组,LED光照功率密度5 mW/cm^2,其中照射时间0 s为对照组,仅接种细胞但不进行光照处理。另设1个空白对照组,即不接种细胞不进行细胞照射。各组处理后24 h均采用CCK8法检测并计算成纤维细胞增殖率。每组设计6个复孔,每组实验重复3次取平均值。结果波长630 nm和810 nm窄带LED在功率密度为5 mW/cm^2,照射时间100~500 s时皆可促进成纤维细胞的增殖,且其促细胞增殖率随照射时间的延长而增加,照射时间达到500 s,能量密度2.5 J/cm^2时,即达到最佳增殖效果。照射时间增至1 000 s,细胞增殖率逐渐进入平台期;照射时间增至2 000 s时,细胞增殖率开始下降,但仍显著高于对照组(P<0.05)。630 nm和810nm两种波长的窄带LED光源在同一光参量下对成纤维细胞的增殖率比较,差异无统计学意义(P>0.05)。结论功率密度5 mW/cm^2,波长630 nm和810 nm窄带LED照射时间100~2 000 s均可促进高糖环境下成纤维细胞的增殖,能量密度2.5 J/cm^2时,细胞增殖作用显著,细胞增殖率不随照射时间的增加而无限增高。Objective To explore the dose-effect relationship of 630 nm and 810 nm narrow-band LED irradiation on the proliferation rate of fibroblasts cultured in high glucose in vitro.Methods The mouse fibroblast cells(L929) cultured in high-glucose DMEM medium and inoculated into 96-well plates at 3 000 cells per well were used as the experimental objects.The experiment was carried out in two groups:the 630 nm LED group and 810 nm LED group.Each group was further divided into 6 LED illumination groups by irradiation lengths(0 s,100 s,200 s,500 s,1000 s and 2 000 s),and the power density was set at 5 mW/cm^2,including a control group in which cells were inoculated but no light treatment was given.Moreover,a blank control group was set,in which neither cell inoculation nor cell irradiation was given.The proliferation rate of fibroblasts was detected with CCK8 method 24 hours after the treatment for the three groups.Each group was designed with 6 duplicate wells,and the average value was repeated 3 times for each experiment.6 duplicate wells were designed for each group and each experiment was repeated 3 times to get the average.Results When the power density was 5 mW/cm^2 and the irradiation time 100-2 000 s,the LED promoted the proliferation of fibroblasts,and its proliferative capacity increased with the extension of the irradiation length.When the irradiation length reached500 s,the best proliferation effect was achieved.When the irradiation length was further extended to 1 000 s,the cell proliferation capacity gradually entered the plateau period.When the irradiation time was prolonged to 2 000 s,the cell proliferation capacity began to decrease slowly,but it was still higher than that of the control group,which was statistically different from that of the blank control group(P<0.05).There was no significant difference between 630 nm and 810 nm narrow-band LED light sources in the proliferation of fibroblasts under the same light parameters(P>0.05).Conclusions Under the parameter conditions used in this experiment 630 n
