摘  要:为了进一步研究森林革蜱钙网蛋白(DsCRT)功能,试验应用PCR方法和生物信息学分析软件预测森林革蜱钙网蛋白基因的结构和功能。通过GenBank比对DsCRT基因和氨基酸序列;利用生物信息学软件对序列进行预测和分析。结果表明:PCR扩增的DsCRT序列长度为933 bp,截短序列编码311个氨基酸,蛋白质分子质量约为35.88 ku,理论等电点约为4.74,亲水性较高;Ds CRT具有良好的抗原表位。系统进化树显示DsCRT基因与变异革蜱和安氏革蜱的亲缘关系最近。说明该蛋白具有较高的免疫原性,且与同属其他种的钙网蛋白具有较高同源性测序获得序列信息为进一步分析DsCRT蛋白的功能提供参考。To further enhance knowledge about calreticulin of D.silvarum,PCR and bioinformatics analysis were used to predict the structure and function of putative D.silvarum calreticulin(DsCRT).The results showed that the length of the truncated DsCRT sequence amplified by PCR was 933 bp,encoding 311 amino acids.The molecular weight of the predicted protein was about 35.88 ku.The theoretical isoelectric point was about 4.74,and the hydrophilicity was high.The DsCRT protein has good antigenicity by prediction.The phylogenetic tree showed that the DsCRT gene was closely related to that of D.variabilis and D.occidentalism,while a little distant from Hyaloma and Rhipicephalus.The sequence information obtained in the study provided a reference for further analysis on the function of D.silvarum calreticulin.
