摘 要:铁死亡是新近发现的一种程序性死亡形式,与凋亡、坏死、自噬等死亡程序不同,其病理特征主要表现为铁离子依赖性脂质过氧化物的蓄积,典型的形态学表现为细胞容积皱缩、线粒体膜面积增加等。铁死亡的机制可能主要与脂质及脂质过氧化物蓄积、氨基酸抗氧化系统失衡、铁离子代谢紊乱等有关,其中甲羟戊酸信号、转硫信号、热激蛋白信号可能是铁死亡程序的重要途径。铁死亡与多种疾病的发生密切相关,但其具体机制尚未完全阐明。了解铁死亡基本内容、其潜在机制和信号通路可为相应疾病诊疗提供方向。Ferroptosis is a newly discovered form of programmed death,which is different from apoptosis,necrosis,autophagy and other death procedures.Its pathological features are mainly the accumulation of iron-ion-dependent lipid peroxides,and the typical morphological manifestations are cell volume shrinkage and mitochondrial membrane area increase.The mechanism of ferroptosis may be mainly related to lipid and lipid peroxides accumulation,imbalance of amino acid antioxidant system,and disturbance of iron ion metabolism,among which,mevalonate signal,sulfur transfer signal and heat shock protein signal may be the important pathway.Ferroptosis is closely related to the occurrence of many diseases,but the specific mechanisms have not been fully elucidated.Understanding the basic contents of ferroptosis,its underlying mechanism and signaling pathway can provide certain directions for the diagnosis and treatment of the associated diseases.
