摘 要:为了解广西猪群猪圆环病毒3型(PCV3)的遗传变异情况,以PCV3阳性DNA为模板,利用PCR的方法分2段扩增PCV3全基因序列并进行拼接,获得10株PCV3全基因组序列,全长均为2000 bp。10株广西PCV3全基因组序列之间的核苷酸同源性为98.6%~99.8%。与国内其他毒株的同源性为97.9%~99.8%;与国外其他毒株的同源性为98.8%~99.7%。10株广西PCV3 Cap基因的大小均为645 bp,编码214 aa。PCV3 Cap基因比较保守,10株广西PCV3 Cap基因与参考株相比,仅存在散在的突变位点。基于PCV3全基因和Cap基因构建的遗传进化树显示,两者结果基本相同,10株广西PCV3处于3a和3b两个亚群。本试验表明,广西地区猪群中流行的PCV3至少存在2个亚群,不同亚群PCV3毒株的致病性差异还有待进一步的研究。In order to understand the genetic variation of porcine circovirus type 3(PCV3)in Guangxi pig herds,PCV3 positive DNA was used as a template to amplify PCV3 genome sequence in two stages by PCR.Ten PCV3 genome sequences were obtained by sequence splicing.The total length of 10 PCV3 strains was all 2000 bp.The nucleotide homology among 10 PCV3 genome sequences was 98.6%~99.8%.The homology with other representative strains in China was 97.9%~99.8%,and was 98.8%~99.7%with other representative strains abroad.The Cap gene of 10 PCV3 strains was 645 bp in size,encoding 214 amino acids.The Cap gene of PCV3 was relatively conserved.Compared with the reference strains,there were only scattered mutation sites in Cap gene of 10 Guangxi PCV3 strains.The phylogenetic trees constructed based on PCV3 genome and Cap gene showed that the results were basically the same.The 10 Guangxi PCV3 were in the subgroups of 3a and 3b.This study showed that there are at least two subgroups of PCV3 prevalent in Guangxi pig herds,but the pathogenicity differences of PCV3 strains among different subgroups need to be further studied.
