摘  要:目的研究右美托咪定对胶质瘤细胞U251体外恶性生长和侵袭的影响和机制。方法以胶质瘤细胞U251作为体外研究对象,右美托咪定处理U251细胞,四甲基偶氮唑蓝(MTT)方法检测细胞增殖情况,Transwell小室测定细胞侵袭和迁移变化,Westernblot检测细胞中基质金属蛋白酶2(MMP-2)、基质金属蛋白酶9(MMIP-9)和p-AKT蛋白表达。用AKT信号激活剂和右美托咪定联合处理胶质瘤细胞,检测细胞增殖、侵袭和迁移能力。结果右美托咪定可降低胶质瘤细胞增殖、侵袭及迁移能力,抑制MMP-2、MMP-9、p-AKT蛋白表达;AKT信号激活剂处理可逆转右美托咪定对胶质瘤细胞增殖、侵袭和迁移能力的影响。结论右美托咪定通过抑制AKT信号通路阻碍胶质瘤细胞U251体外恶性生长和侵袭。OBJECTIVE To study the effect and mechanism of dexmedetomidine on the malignant growth and invasion of gli-oma cell line U251 in vitro.METHODS Using glioma cell U251 as an in ritro study,treated with dexmedetomidine,MTT as-say was used to measure the cell proliferation,Transwell charmbers was used to measure the changes in cell invasion and migra-tion,and the expressions of MMP-2,MMP-9 and p-AKT protein in cells were detected by W estern blot.Clioma cells were trea-ted with AKT signaling activator and dexmedetomidine,and cell proliferaion,invasion and migration were observed.RESULTS Dexmedetomidine can reduce the proliferation,invasion and migration of glioma cells and inhibit the expression of MMP-2,MMP-9 and p-AKT proteins.AKT signaling activator lreatment reversed the flect of dexmedetomidine on the proliferation,inva-sion and migration of glioma cells.CONCLUSION Dexmedetomidine blocks malignant growth and invasion of glioma cell U251 in vitro by inhibiting AKT signaling.
