摘 要:目的研究ATP生物荧光法检测气溶胶细菌浓度的方法,为其应用提供科学数据。方法采用人工气溶胶方法,对ATP生物荧光法检测不同试验菌人工气溶胶细菌浓度进行观察,同时与活菌计数法作比较。结果建立了粘质沙雷菌、金黄色葡萄球菌、大肠埃希菌及其混合菌液浓度与ATP发光值的关系曲线,线性回归分析得出各曲线的R2值均>0.9,显示在103~109cfu/m L范围内细菌浓度与生物发光强度有明显的正相关性。混合菌液气溶胶的模拟试验检测结果显示,ATP生物荧光法和平板计数法在气溶胶浓度较高时有较好的相关性,R2值为0.8613。ATP生物荧光法检测结果受细菌气溶胶浓度、不同的采样器和采样方式等因素的影响。结论ATP生物荧光法快速检测空气中细菌浓度的方法具有可行性,但结果会受到诸多因素的影响,尚需要进一步研究解决。Objective To establish the ATP bioluminescence method for detecting microbial aerosol concentration,and to provide scientific data for its application.Methods In artificial aerosol method,the artificial aerosol bacteria concentration of different test bacteria was detected by ATP bioluminescence method,which was compared with live bacteria counting method at the same time.Results The relationship curve between the concentration of Serratia marcescens,Staphylococcus aureus,Escherichia coli and their mixed bacterial solution and ATP luminescence value was established.The linear regression analysis showed that the R2 value of each curve was greater than 0.9,and the bacterial concentration in the range of 103~109 cfu/m L had a significant positive correlation with the bioluminescence intensity.The detection results of simulation experiments of mixed bacterial aerosols showed that ATP bioluminescence method had a better correlation with the plate count method when the aerosol concentration was higher,and the R2 value was 0.8613.The results of ATP biofluorescence detection were affected by factors such as microbial aerosol concentration,different samplers and sampling methods.Conclusion The rapid detection of microbial aerosol concentration based on the ATP bioluminescence method is feasible,but the result will be affected by many factors,and further research and solution are needed.
