摘　　要：[目的]分析脂多糖诱导小胶质细胞中炎症相关的长链非编码RNA (long non-coding RNA, lncRNA)表达。[方法]将小鼠小胶质细胞(BV2)样本随机分为两组,脂多糖(lipopolysaccharide, LPS) 1ug/ml处理24 h后细胞组为LPS组,未做LPS干预细胞为对照组。通过生物信息学分析后,实时荧光定量PCR (Quantitative Real-time PCR, qPCR)进一步验证LPS诱导后BV2的lncRNA与炎症的关系。[结果]在高通量测序中选取炎症相关差异表达明显的9个lncRNA,qPCR验证后发现AC005899.1、GPATCH8、VPS41与高通量测序中的表达趋势一致。[结论]多个lncRNA可能参与体外小胶质细胞的炎症反应过程。这些结果为小胶质细胞通过lncRNA参与神经病理性疼痛提供相关信息。[Objective]Analysis of lipopolysaccharide-induced inflammation-associated long non-coding RNA(lncRNA)expression in microglia.[Methods]The microglia(BV2)cells were divided into two groups.Of them,the cells that were activated by lipopolysaccharide(LPS)1 ug/ml for 24 hours termed as the LPS group,while those were cultured in normal condition without treatment of LPS named as the control group.The relationship between the microglia cell activation and inflammation was assessed by bioinformatics analysis and qPCR assay.[Results]In term of the high-throughput sequencing,9 lncRNAs relative to inflammation were identified in the LPS group,which was obviously different with the control group.As results of qPCR assay,the lncRNAs,including AC005899.1,PATCH8,VPS41 and so on,were found in the LPS group,which was significant different in expression level between the LPS group and the control group(P<0.05),whereas was consistent with the expression trend by high-throughput sequencing.[Conclusion]Many lncRNAs involve in the inflammatory process of BV2 microglia cells in vitro.These results indicate that microglia can participate in neuropathic pain through lncRNA.
