摘　　要：目的分析心房肌超极化激活环核苷酸门控阳离子通道2(HCN2)及超极化激活环核苷酸门控阳离子通道4(HCN4)基因在心房颤动大鼠心肌中的表达,观察青山健心片对HCN2、HCN4 mRNA及蛋白表达的干预作用,揭示其治疗心房颤动的机制。方法取阵发性心房颤动大鼠心房组织,采用蛋白免疫印迹法(Western-Blot)及荧光定量聚合酶链式反应(RT-PCR)法检测HCN2、HCN4的表达,分析青山健心片的干预作用。结果阵发性心房颤动大鼠HCN2、HCN4 mRNA及蛋白表达量均高于对照组,青山健心片及美托洛尔均能降低HCN2、HCN4 mRNA及蛋白表达。结论青山健心片通过下调HCN2、HCN4表达,影响超极化激活阳离子电流(If),从而减少心房颤动发作。Objective To analyze the synergistic expression of HCN2 and HCN4 genes in the myocardium of rats with atrial fibrillation,and observe the effect of Qingshan Jianxin Tablet on the expression of HCN2,HCN4,reveal the mechanism of Qingshan Jianxin Tablet in the treatment of atrial fibrillation.Methods The expression of HCN2 and HCN4 were detected by Western-blot and RT-PCR in atrial tissue of rats with paroxysmal atrial fibrillation.Results The results of Western-blot and RT-PCR showed that the mRNA and protein expression levels of HCN2 and HCN4 in the atrial fibrillation group were higher than those in control group.Both Qingshan Jianxin Tablet and metoprolol could reduce mRNA and protein expression.Conclusion Qingshan Jianxin Tablet can reduce the incidence of atrial fibrillation by down-regulating the expression of HCN2 and HCN4 and affecting If current.
