摘  要:目的通过检测泡球蚴感染小鼠小肠白细胞介素-25(IL-25)及其mRNA表达水平的变化,探讨其在泡球蚴感染中的作用。方法将64只雌性BALB/c小鼠随机分为对照组与模型组。取泡球蚴保种鼠肝脏囊泡中的原头蚴,处理后通过腹腔注射建立泡球蚴小鼠动物模型,对照组注射生理盐水。在模型建立后,分别于第2天、第8天、第14天以及第30天各取8只感染的小鼠处死,在无菌条件下收集小肠组织,采用免疫组织化学法检测IL-25在小肠中的表达水平,采用qRT-PCR检测IL-25 mRNA相对表达量。结果在免疫组化实验中,与对照组比较,模型组小鼠小肠中IL-25表达水平在感染第2天后升高(P<0.05),感染第8天后继续上升,与对照组相比差异具有统计学意义(P<0.01),感染第14天后再次出现明显上升(P<0.01),在第30天时IL-25的表达到达顶峰(P<0.01)。qRT-PCR实验中,小鼠小肠组织IL-25的mRNA表达水平与免疫组化趋势结果一致。结论小鼠感染泡球蚴后,小肠组织中IL-25的表达在感染早中期呈逐渐上升的趋势,提示小肠组织中IL-25是泡球蚴感染早中期的重要参与细胞因子之一。Objective To investigate the changes of IL-25 and its mRNA expression level in the small intestine of mice infected with Echinococcus multilocularis,and explore its role in the infection of alveolar echinococcosis.Methods 64 female BALB/c mice were prepared and randomly divided into a control group and a model group.The E.multilocularis infected model were established by intraperitoneal injection of protoscoleces of E.multilocularis.And the control group was injected with saline.After establishment of the mice model,at each time points(2nd,8th,14th,and 30th days),8 infected mice were sacrificed and their small intestinal tissues were collected under sterile conditions,and the IL-25 was detected by immunohistochemistry.The expression level of IL-25 in the small intestine was determined by qRT-PCR.Results The immunohistochemical results showed that,compared with the control group,the expression level of IL-25 in the small intestine of the model group was increased at the 2nd day of infection(P<0.05),and was continually increased at the 8th day of infection(P<0.01).At the 14th day of infection,the expression of IL-25 was increased again(P<0.01).At the 30th day of infection,the expression of IL-25 was reached its peak level(P<0.01).The qRT-PCR results showed that,the mRNA expression of IL-25 in mouse small intestine was consistent with the results of immunohistochemistry.Conclusion After infection of alveolar echinococcosis,the expression of IL-25 in the small intestine was gradually increased atthe early and middle stages of infection.It suggested that IL-25 in the small intestine is one of the important cytokines involved in the early and middle stages of alveolar echinococcosis infection.
