摘 要:目的对河北省鼠疫菌株进行规律聚集间隔短回文重复序列CRISPR)基因分型。方法利用聚合酶链式反应(PCR),采用3对CRISPR引物,对河北省分离的128株鼠疫菌进行扩增,测定扩增产物核酸序列并进行分析,确定河北省菌株CRISPR基因型。结果128株鼠疫菌在3个CRISPR位点上有8种间隔序列,包括al、a2、a3、b1、b2、c1、c2、c3,分为2个基因型。118株鼠疫菌株为Cb2型,另外10株鼠疫菌均为新基因型(命名为Cc△1型)。结论通过CRISPR分析显示河北省鼠疫菌株遗传进化比较稳定,建立了CRISPR基因库,为鼠疫菌种群遗传进化和鼠疫防控的研究提供参考。Objective To understand the CRISPR genotyps of Yersinia pestis strains in Hebei Province.Methods A total of 128 strains of Yersinia pestis isolated from Hebei Province were amplified by using 3 pairs of CRISPR primers,the nucleic acid sequences of the amplified products were determined and analyzed,and the CRISPR genotypes of the strains were determined by cluster analysis using BioNumerics software.Results Eight spacers based on 3 CRISPR loci,including AL,A2,A3,B1,B,C1,C2 and C3 were found from the 128 strains of Yersinia pestis.There were two genotypes,Cb2 and Cc△1(new genotype).Of the 128 Yersinia pestis strains tested,118 were Cb2,and the other 10 were Cc△1.Conclusion CRISPR analysis showed that the genetic evolution of Yersinia pestis strains in Hebei Province was stable,and the establishment of CRISPR Gene Bank provided reference for the study of genetic evolution of Yersinia pestis population and prevention and control of Yersinia pestis.
