多花黄精丛生芽的诱导与增殖条件

摘　　要：以多花黄精种子萌发的初生根茎为外植体,研究细胞分裂素不同种类(6-BA、KT、ZT)及不同浓度(2.0、3.0、4.0 mg·L-1)与NAA和GA3组合对丛生芽诱导效率的影响,并探讨不同基本培养基(MS、White、B5、N6)、不同植物生长调节物质(细胞分裂素类6-BA、TDZ、ZT,生长素类NAA、IAA、2,4-D)、p H值、温度及光照强度对丛生芽增殖的影响。结果表明:多花黄精丛生芽诱导的适宜培养基为MS+6-BA 4.0 mg·L-1+NAA0.25 mg·L-1+GA30.5 mg·L-1+蔗糖30 g·L-1+琼脂5 g·L-1,诱导率为85.19%;一定浓度的6-BA、NAA和IAA组合可有效促进多花黄精丛生芽的增殖;适于丛生芽增殖的培养基为MS+6-BA 3.0 mg·L-1+NAA0.4mg·L-1+IAA0.1mg·L-1+蔗糖30g·L-1+琼脂5g·L-1,p H值为5.8,温度为25°C,光照强度为25~37.5μmol·m-2·s-1,培养30d后增殖倍数可达13.44。The primary rhizomes of Polygonatum cyrtonema from seed germination were used as explants.Cluster buds were induced by different kinds of cytokinins(6-BA,KT and ZT),concentration(2.0,3.0 and 4.0 mg·L-1)and their combination with NAA and GA3.Meanwhile,the effects of cluster buds proliferation were also discussed by different basic media(MS,White,B5 and N6),plant growth regulators(6-BA,TDZ,ZT,NAA,IAA and 2,4-D),p H value,temperature and illumination intensity treatments.The results showed that MS+6-BA 4.0 mg·L-1+NAA 0.25 mg·L-1+GA30.5 mg·L-1+sucrose 30 g·L-1+agar 5 g·L-1 was the suitable medium for inducing cluster buds of P.cyrtonema,and the induction rate was 85.19%.The combination of 6-BA,NAA and IAA at a certain concentration could effectively promote the proliferation of cluster buds.The suitable medium for cluster buds proliferation was MS+6-BA 3.0 mg·L-1+NAA 0.4 mg·L-1+IAA 0.1 mg·L-1+sucrose 30 g·L-1+agar 5 g·L-1,p H value was 5.8,temperature was 25°C,illumination intensity was 25 to 37.5μmol·m-2·s-1,the multiplication multiples of cluster buds reached 13.44 after 30 days culture.
