摘 要:目的研究3D打印可控式张力带技术对皮肤缺损动物模型中创面愈合和皮肤再生的影响及其机制。方法取6月龄、体质量2~3kg的新西兰大白兔构建皮肤缺损动物模型,以不植入3D打印可控式张力带装置为对照组,植入3D打印可控式张力带装置为实验组,分别是无张力带组、常规张力带组和快速张力带组。各组于植入手术后3、7和14天检测大白兔体温和体重变化,同时采集术后14天时大白兔的皮肤组织,分析创面收缩率,采用苏木精-伊红(HE)染色,观察皮肤组织的病理学变化,通过细胞角蛋白(pan-cytokeratin)、血小板-内皮细胞黏附分子(CD31)和血管内皮生长因子(VEGF)免疫组织化学染色,观察表皮生长和血管形成的过程。结果在植入3D打印可控式张力带装置后,对照组和实验组体温和体质量差异无统计学意义,创面愈合观察和收缩率结果显示,与对照组比较,无张力带组无显著变化,常规张力带组和快速张力带组创面干燥,结痂和愈合速度快,创面收缩率显著高于对照组(P<0.05),其中常规张力带组创面收缩率最高。HE染色显示,对照组和实验组全层皮肤未见明显的病理学变化,免疫组织化学染色显示,与对照组比较,无张力带组无显著变化,常规张力带组和快速张力带组增加了全层皮肤表皮厚度,上调了CD31和VEGF的表达,其中常规张力带组最为显著(P<0.05)。结论在皮肤缺损动物模型中,3D打印可控式张力带技术可通过控制张力大小促进创面愈合和皮肤再生,其中常规张力带组作用最显著,其机制可能是通过刺激表皮生长和上调全层皮肤组织的CD31和VEGF的表达加速新血管的形成来促进皮肤再生。Objective To study the effects of 3D printed controllable tension band technology on wound healing and skin regeneration in skin defect animal models and its mechanism.Methods New Zealand white rabbits aged 6 months and weighing 2-3kg were used to construct an animal model of skin defect.The control group was implanted without a 3D-printed controllable tension band device and the experimental group was implanted with a 3D-printed controllable tension belt device,including tension-free band group,conventional tension band group and fast tension band group.Changes in body temperature and body weight of white rabbits were detected at 3,7,and 14 days after implantation in each group.At the same time,skin tissues of the white rabbit was collected at 14 days after surgery,the wound shrinkage was analyzed,Hematoxylin-Eosin(HE)staining was used to observe the pathological changes of skin tissue.Immunohistochemical staining of pan-cytokeratin,platelet-endothelial cell adhesion molecule(CD31)and vascular endothelial growth factor(VEGF)was used to observe the process of epidermal growth and angiogenesis.Results After implanting the 3D printed controllable tension band device,there was no significant difference in body temperature and weight between the control group and the experimental group.The wound healing observation and shrinkage results showed that compared with the control group,there was no significant change in the tension-free band group.The conventional tension band group and the fast tension band group wound healing were dry,crusting and healing were fast,and the wound shrinkage rate was significantly higher than that of the control group(P<0.05).The conventional tension band group had the highest wound shrinkage rate.HE staining showed no significant pathological changes in the full-thickness skin of the control and experimental groups.Immunohistochemical staining showed that compared with the control group,there was no significant change in the tension-free band group.The conventional tension band group and t
