摘 要:以‘吉椒15号’和‘吉椒16号’辣椒及其亲本为研究材料,筛选快速鉴定其种子纯度的SSR分子标记。结果发现,72对候选SSR引物中,有6对在杂交种及亲本之间具有多态性且重复性好的共显性标记,其中SSR11、SSR16、SSR34、SSR48及SSR52分别在‘吉椒15号’及其亲本间具有多态性,SSR11、SSR14、SSR16及SSR52分别在‘吉椒16号’及其亲本间具有多态性,上述6对SSR引物可以作为鉴定种子纯度的分子标记。研究结果表明,采用单个SSR标记或2~3个组合标记可以快速鉴定出‘吉椒15号’和‘吉椒16号’辣椒种子中混杂的母本或异形株,SSR分子标记鉴定体系的初步建立可以为辣椒种子纯度的鉴定工作提供便利。‘Jilin pepper 15’and‘Jilin pepper16’together with their parents were used as the materials in this study to establish their seed purity identification system using SSR markers.Among the 72 pairs of candidate SSR primers,there were 6 pairs of primers exhibiting polymorphism and good repeatability between the hybrids and their parents.Markers of SSR11,SSR16,SSR34,SSR48 and SSR52 were polymorphic between‘Jilin pepper 15’and its parents,whereas SSR11,SSR14,SSR16 and SSR52 were polymorphic between‘Jilin pepper 16’and its parents.These markers were suitable to be used for seed purity testing.The results showed that single SSR marker or combination of 2~3 markers could quickly identify the hybrid female parents or heterotypic plants in the seeds of‘Jilin pepper 15’and‘Jilin pepper 16’.The identified SSR markers provided convenience for the identification of pepper seed purity.
