通痹降浊汤治疗高尿酸血症大鼠的药效学观察

摘　　要：目的观察通痹降浊汤对高尿酸血症大鼠模型的药效学影响。方法通痹降浊汤由北京中医药大学深圳医院制剂室制备,将60只雄性SD大鼠按体质量随机分为空白对照组、模型组、低剂量中药组、高剂量中药组和别嘌醇组,每组12只。实验周期14d,药物组大鼠预先灌胃给药7d,第8天开始使用酵母膏15g/kg+腺嘌呤100mg/kg灌胃,连续7d,最后1d灌胃氧嗪酸钾300mg/kg造模。检测大鼠血尿酸、尿素氮,苏木素-伊红染色(HE染色)法检测肾组织病理。结果与空白对照组相比,模型组大鼠血清尿酸、尿素氮含量显著升高,且肾脏病理组织有一定程度损害,两个剂量中药均具有降低高尿酸血症大鼠血清尿酸的作用,且可显著降低大鼠血清尿素氮水平,减轻肾脏病理改变,其作用无明显剂量依赖性。结论通痹降浊汤对于高尿酸血症大鼠具有明显治疗作用。Objective To observe the pharmacodynamic effects of Tongbi Jiangzhuo Decoction on rats with hyperuricemia.Methods Tongbi Jiangzhuo Decoction was prepared by the preparation room of Shenzhen Hospital Affiliated to Beijing University of Chinese Medicine.60 male SD rats were randomly divided into the control group,model group,low-dose Chinese medicine group,high-dose Chinese medicine group and Allopurinol group according to their body weight,12 animals in each group.The experimental period was 14 days.Rats in the drug groups were administered by intragastric for 7 days in advance.On the 8th day,the rats model of hyperuricemia was established by intragastric yeast extract 15 g/kg and adenine 100 g/kg for 7 consecutive days,and the rats were given potassium oxalate 300 mg/kg once on the last day.Rats were killed one hour after.Serum uric acid(UA)and Serum urea nitrogen(BUN)were measured by kits.Kidney pathology was detected by Hematoxylin-eosin staining(HE staining).Results Compared with the control group,the UA and BUN in the model group were significantly increased and the kidneys showed obvious pathological damages.Compared with the model group,the levels of UA and BUN were significantly reduced in both doses of the Tongbi Jiangzhuo groups.And the kidney pathological damage in hyperuricemia rats was also alleviated.Its effect has no obvious dose-dependent.Conclusion Tongbi Jiangzhuo Decoction has a significant therapeutic effect on rats with hyperuricemia.
