摘 要:建立了一种同时测定黄连中3种原小檗碱型生物碱的高效液相色谱法,该方法可以使黄连中的小檗碱、巴马汀、药根碱得以分离和检测.采用Agilent Zorbax C18(250 mm×4.6 mm,5μm)色谱柱,以乙腈-20 mmol/L磷酸二氢钾缓冲溶液(pH 3.0,含0.2%三乙胺)梯度洗脱,柱温25℃,流速1 mL/min,在波长为265 nm处检测.结果表明,各组分线性关系良好,相关系数R为0.999 5~0.999 6,加标回收率在92.9%~105.9%,检出限0.1~0.45μg/mL.A method based on high-performance liquid chromatography coupled with diode array detection(HPLCDAD)has been developed for simultaneous determination of three protoberberine akaloids(berberine,palmatine and jatrorrhizine)in Rhizoma coptidis. The analysis was performed on a Zorbax SB C18 column with gradient elution of water(0.02 mol·L-1 sodium dihydrogen phosphate,0.2% triethylamine,adjusted to p H 3 with phosphoric acid)and acetonitrile. The column temperature was 25 ℃,flow rate was 1 m L/min,and UV detection was set at 265 nm. All calibration curves showed good linearity with correlation coefficients 0.999 5-0.999 6 and spiked recovery were92.9%-105.9%. Limit of detection was in the range of 0.1-0.45 μg/mL. The results reveal that the established method is suitable for the determinations of berberine,palmatine and jatrorrhizine in R. coptidis.
